摘 要:目的观察依那普利抑制培养的血管平滑肌细胞增殖的机制。方法以组织贴块法行自发性高血压大鼠(spontaneouslyhypertensiverats,SHR)主动脉血管平滑肌细胞的分离培养,并以倒置显微镜α-SM肌动蛋白免疫组化法鉴定;分别采用甲基噻唑基四唑(MTT)法、氚-胸腺嘧啶核苷(3H-TdR)掺入法、荧光染料(FURA-2/AM)标记法。酶底物反应法观察依那普利含药血清对血管平滑肌增殖、细胞内游离钙、蛋白激酶C活性的影响。结果与模型组比较,依那普利含药血清对血管平滑肌增殖、脱氧核糖核酸(DNA)合成、细胞内游离钙、蛋白激酶C活性均有明显的抑制作用(P<0.01)。结论依那普利可能通过降低细胞内游离钙水平及抑制蛋白激酶C活性而发挥抑制血管平滑肌增殖和高血压血管重构的作用。Objectives To study the mechanism of enalapril on cultured vascular smooth muscle cells (VSMC)proliferation. Methods VSMC were isolated from the media of spontaneous hypertensive rats (SHR) aorta obtained by explant method and identified by inverted microscope and a-SM actin immunohistochemical method. The effects of the drug containing serum on VSMC proliferation was observed by MTT method, DNA synthesis by 3H-TDR incorporated method, intracellular free calcium ion[(Ca^2+]i) by FURA 2/AM marking method and protein kinase C (PKC) activity by PcpTag non-radioactive PKC assay. Results Enalapfil had inhibiting effects on VSMC proliferation, DNA synthesis and hyper-VSMC [Ca^2+]i and PKC activity, with better effects than control group. Conclusions Enalapril has marked inhibitory effects on the hyper-VSMC [Ca^2+]i and PKC activity,and hence improve the VSMC proliferation and vascular remodeling.
