摘  要:目的:测定肽类药物曲普瑞林、促黄体素释放激素类似物的分子量及其一级结构。方法:应用电喷雾质谱法(ESI MS)及源内碰撞诱导解离技术对肽类药物曲普瑞林及促黄体素释放激素类似物进行测定,根据其典型碎片离子确证其氨基酸序列及分子量。结果:肽类化合物在正离子模式下可得到较好的质谱信息,2种寡肽均测得其准分子离子峰(M+H)^+及(M+Na)^+信号,分子量分别与理论值一致。利用电喷雾碰撞诱导解离方法,得到一系列典型的 y 及 b 系列碎片离子,从而确证了2种寡肽的一级结构。结论:本研究为测定寡肽的分子量和一级结构提供了一个简便、快速、准确的方法。Objective: To determinate the molecular weight and the first-order structure of triptorelin and luteinizing releasing hormone analog(LRH-A). Method: The molecular weight and amino acid sequence of triptorelin and LRH - A were determined by electrospray ionization mass spectrometry. The y and b series of fragment ions of triptorelin and LHR-A were obtained by in-source collision-induced dissociation(CID). Results: The m/z of quasi - molecular ion(M + H)^+ of triptorelin was 1311.6 and (M + Na)^+ was 1333.5. It means that the molecular weight of triptorelin was 1310.6. The m/z of quasi - molecular ion (M + H)^+ of LRH - A was 1167.6 and (M + Na)^+ was 1189.6. It means that the molecular weight of LRH - A was 1166.6. The y and b series of fragment ions were obtained by adjusting the fragmentor voltage. The first order structure of triptorelin and LHR - A were confirmed. Conclusion: A sensitive, rapid and accurate method was established for the determination of oligopeptide.
