摘 要:目的:测定中药柴胡(BupleurumChineseDC,BCDC)提取物对人肝癌BEL-7402细胞内游离钙离子浓度([Ca2+]i)和长春新碱(Vincristine,VCR)浓度的影响,探索柴胡提取物逆转BEL-7402细胞多药耐药的机制。方法:以Fura2/AM为细胞内钙离子的荧光指示剂,用双波长荧光分光光度计分别测定不同条件下的BEL7402细胞内[Ca2+]i。高效液相色谱法测定BCDC作用于BEL-7402后细胞内VCR蓄积情况的变化。结果:柴胡提取物可使人肝癌细胞BEL-7402细胞内游离钙离子浓度下降(P<0.001),可使细胞内VCR浓度升高(P<0.01)。结论:柴胡提取物可以增加VCR在BEL7402细胞内的积聚浓度,部分逆转BEL7402细胞的多药耐药(multidrugresistance,MDR)。钙离子通道阻滞作用可能为柴胡提取物逆转BEL7402细胞多药耐药的机制之一。Objective: To explore the mechanism by which Bupleurum Chinese DC (BCDC) reverses the mutidrug resistance (MDR) of human hepatoma BEL-7402 ceils by measuring intraeellular concentrations of free calcium and vincristine (VCR). Methods: Intracellular free calcium concentration ([Ca^2+ ]i) in human hepatoma BEL-7402 cells of different groups was measured by double wavelength speetrofluorometer with Fura-2/AM as the fluorescence indicator. Cellular VCR concentration after BCDC treatment was examined by high performance liquid chromatography (HPLC). Results:Cellular VCR concentration in BEL-7402 cells was increased after treatment with 390 μg/ mL BCDC and 5μg/mL verapamil (VRP) compared with control,respectively (P〈0.01 ). VRP and 390μg/mL BCDC increased intraccllalar VCR from (57. 212±0.032) pg/μL up to (60. 145~0. 028) pg/μL and (59. 196~0. 045)pg/bd.,respectively. BCDC reduced [Ca^2+]i in human hepatoma BEL-7402 cells(P〈0.001). Condusion:BCDC increased intraeellular VCR concentration in BEL-7402 cells indicating that BCDC partly reversed MDR of BEL-7402 cells. Blockade of calcium channels may be one of the mechanisms by which BCDC reverses the MDR of human hepatoma BEL- 7402 cells.
