摘  要:建立了一种快速检测氨基甲酸酯类农药西维因痕量残留的新方法,即膜载体酶标记渗透式快速检测技术。以带正电尼龙膜(M illipore)为固相载体,辣根过氧化酶标记的西维因半抗原为检测标记物,采用直接竞争酶联免疫吸附方式对农产品中的痕量西维因残留进行快速检测。该方法完成整个检测过程只需5m in,对西维因的检出限为10μg/L,通过高效液相色谱法(HPLC)验证了该检测方法的有效性,并经试验证明了方法的稳定性。A rapid and sensitive osmotic immunoassay for carbaryl was developed. The assay was based on the competitive principle and used anti-carbaryl polyclonal antibody immobilized on Nylon^+ membrane. Hapten of carbaryl labelled with horseradish peroxidase (HRP) was used as tracer. The experimental condition were as follows: anti-carbaryl antibody coatedat 1.0 μg/L, 600 μL carbaryl (or sample extract containing carbaryl) mixed with 100 μL carbaryl hapten-HRP, 120 μL mixture was added on the zone that coated with antibody. After the liquid filtered the membrane, it was washed to remove unreacted chemicals. Substrate solution was uniformly added on the membrane. The total assay time was less than 5 minutes and the visual detection limit was 10 μg/L. The developed assay has good stability. By comparing the results of samples obtained by high performance liquid chromatography and osmotic immunoassay, the correlation between the two methods was good (χ^2 =4.0103, p 〈0.01).
