摘  要:目的利用人胃癌BGC823细胞体外培养实验,观察不同浓度ω3多不饱和脂肪酸(EPA、DHA)对肿瘤细胞周期及凋亡的影响。方法采用MTT法、凋亡形态学检查和流式细胞检测肿瘤细胞周期及凋亡情况。结果30和45μg/ml的EPA或DHA可显著降低肿瘤细胞存活率(P<0.001);Hoechst33258染色和透射电镜可见细胞凋亡现象;流式细胞检测bcl2基因蛋白表达明显下降(P<0.05),G0/G1细胞明显增加,G2/M和S期细胞明显减少(P<0.05)。结论ω3多不饱和脂肪酸可诱导人胃癌BGC823细胞凋亡和阻滞细胞在G0/G1期,细胞增殖受到抑制。To assess the effects of the ω-3 polyunsaturated fatty acids ( different concentration of EPA and DHA) on cell cycle and apoptosis of human gastric cancer cell line BGC-823 in vitro. Methods Cell cycle and apoptosis of the cells were determined by MTT assay, morphology, and flow cytometry. Results After treatment with 30 and 45μg/ml of EPA or DHA, the cell viability significantly decreased ( P 〈 0. 001 ). Apoptosis on the treated cells was observed with Hoechst33258 staining and transmission electron microscope. Bcl-2 protein expression significantly decreased and cells at G2/M phase and S phase were significantly decreased ( P 〈 0. 05 ), whereas G0/G1 phase was increased ( P 〈 0. 05 ). Conclusion ω-3 polyunsaturated fatty acids can induce apoptosis of human gastric cancer cell line BGC-823 cells and inhibit the growth of the cultured cells with them accumulating in G0/G1 phase.
