摘 要:目的探讨 AD 患者脑神经元凋亡的可能机制。方法将死后6h 以内取材的 AD患者和非 AD 患者脑颞叶组织分为两组,每组分别取3例,匀浆后用 Lowry 法进行蛋白定量,将总蛋白量相等的样品进行免疫组织化学双染色,Western 印迹法分析。结果 AD 患者和对照组非 AD患者脑组织切片中均存在凋亡细胞,但 AD 患者脑组织切片中单位面积凋亡细胞增加。AD 患者颞叶组织神经生长因子(NGF)、受体酪氨酸激酶 A(TrkA)、磷酸化的 cAMP 反应元件结合蛋白(P-CREB)、抗凋亡蛋白 Bcl-2表达减少,凋亡前期蛋白 Bax 及 caspase-6表达增加。结论 AD 患者颞叶存在神经元调亡,其存活信号转导通路受到损害;AD 患者颚叶神经营养因子及其受体表达减少。神经营养因子表达的减少以及存活信号转导通路的损害可能是导致 AD 脑神经元凋亡的原因之一。Objective To explore the mechanisms of brain neuron loss and apoptosis, which were primary characteristics in Alzheimer disease(AD). Methods (1)Immunohistochemistry was used for double staining with TUNEL and NSE antibodies. (2)Temporal tissue was taken from 3 cases with AD and 3 cases without AD. Protein estimation was done by Lowry method. Equal amount of protein was taken from each specimen and electrophoresis was done and analyzed by Western blot. Color development was done by alkaline phosphatase method or Luminol Reagent. Results ( 1 ) TUNEL positive brain neurons occurred in AD and non-AD, but the number in the former was more than the one in the latter. (2)AD temporal tissue showed diminished expressions of NGF, apoptosis -related protein Bcl-2 and P-CREB, and increased expressions of apoptosis-related protein BAX and caspase-6. Conclusions Expression of NGF diminished in AD brains. This suggests the diminished expression of NGF and impairment of neuron survival signal transduction pathway may be one of the reason of apoptosis in AD brains.
