摘 要:目的:探讨雏鸡高度近视眼眼球后壁组织血管活性肠肽(vasoactiveintestinalpeptide,VIP)mRNA的动态表达及意义。方法:选择出生1d来亨雏鸡共30只,15只右眼遮盖作为实验组,分别持续遮盖1,2和4wk;另15只双眼均未遮盖作为正常对照组。两组均采用检影验光检测屈光度;眼科A超测定活体眼轴长度;对两组3个时间段眼球后壁行RT-PCR检测VIPmRNA的动态表达。结果:实验组由实验前远视眼快速演变为高度近视眼,并随遮盖时间延长,近视度数加深,眼轴延长;两组眼球后壁均有VIPmRNA阳性表达;随出生时间延长两组VIPmRNA表达均逐渐增强。与对照组相比,实验组VIPmRNA表达明显上调(P<0.01)。结论:形觉剥夺可导致高度近视眼;高度近视眼较正常眼VIPmRNA表达明显上调。VIP可能参与了近视眼形成与发展的调控。AIM: To investigate the dynamic expression and significance of vasoactive intestinal peptide (VIP) mRNA on posterior eyeball in high myopia. METHODS: 30 yellow health leghorn chickens of one-day-old were chosen. 15 chickens were experimental animals using the mebhod of form deprivation with the right eyes covered continuously for 1,2 and 4wk separately. The other 15 chickens not covered were regarded as control group. Both groups were detected diopter using retinoscopic refraction, determined eyeball axis using ophthalmology ultra-A, and investigated the dynamic expression of VIPmRNA on posterior eyeball at three stages by RT-PCR. RESULTS: The experimental eyes were changed from hypermetropia at pre-experiment to high myopia quickly during experiment period, and the diopter degrees were deeper and eyeball axis were longer accompanying with the period elongation of form deprivation. Both groups had the expression of VIPmRNA and increased following the age. Compared with the control, the expression of VIPmRNA of experimental groups were significant up-regulatod (t=15.892,16.278,21.480, P〈0.01). CONCLUTION: Form deprivation can succeed in setting up myopia animal's model, cause the eyes axis to lengthen , induce to axis myopia; The diopter degrees were deeper accompanying with the period elongation of form deprivation. Form deprivation on chickens made the expression of VIPmRNA on posterior eyeball up-regulate significantly. VIP may play an important role on the formation and development of myopia.
