摘  要:目的建立一种与人冠状动脉旁路移植手术(coronary artery bypass graft,CABO)后大隐静脉桥狭窄病理过程相似的动物模型。方法20~25k普通长白猪8只.后腿外侧纵切口,“no-touch”技术剥取大隐静脉3cm,与颈总动脉进行端端吻合。术后口服肠溶阿司匹林150mg·d^-1抗凝。术后28d取下静脉桥,采用计算机图像处理系统分析新内膜厚度、新内膜面积,(新内膜+中膜)面积(I/M)。结果肉眼观察,8只猪大隐静脉桥管壁均增厚,管腔狭窄;光镜下见静脉桥新内膜形成,新内膜厚度为(0.5009±0.1355)mm,I/M为(0.4651±0.0512)。结论猪大隐静脉-颈总动脉静脉桥模型可成功模拟人CABG术后大隐静脉桥的病理变化。Objective To constitute a vein graft model that can mimic the pathology of human saphenous vein graft. Methods Studies were performed by using "white race" pigs ( n = 8), weight 20 - 25 kg. Animals were subjected to premedication, anesthesia. A longitudinal incision was made on the outer aspect of the hind leg to ex "no pose and remove approximately 3 cm in length of the long saphenous vein with a touch" technique. A 3 cm segment of one common carotid artery was isolated through a longitudinal neck incision, and both cut ends were then beveled to about 45 degree. One end of a segment of reversed saphenous vein was similarly beveled and then anastomosed to the carotid artery by an end-to-end anastomotic technique under optical magnification with continuous 8-0 polyprolene suture. The second anastomosis was performed similarly. Proximal anastomosis was always done first.The graft was de-aired through the suture line before the suture of the second anastomosis was tied. The animals were fed a normal chow diet and aspirin, 150 mg·d^-1 for 28 days. Grafts were then removed, fixed with the use of 10% buffered formal saline solution, and then processed for wax embedding. Transverse (5μm) sections were stained with haematoxylin-eosine, and examined under a light microscope. Vessel wall dimensions were measured by computer-aided planimentry. Results All the grafts were patent, in that all saphenous vein graft wall thickening and lumen stenosis were examined by baked-eye. Neointimal proliferation was examined under a light microscope. Neointimal thickening was equivalent to (0. 500 9 ± 0.135 5) mm, neointimal area divided by neointimal area and medial area was equivalent to 0.465 1 ± 0.051 2. Conclusion We constituted a porcine saphenous vein into tarotid artery interposition grafting medel which can mimic the pathology of human saphenous vein graft.
