摘 要:目的研究IL-6诱导相关新基因LX3在真核细胞内的表达定位。方法构建真核融合表达载体pEGFP-N1/LX3,运用Lipofectin转染293T和NIH3T3细胞。共聚焦荧光显微镜检测GFP荧光,以确定IL-6诱导相关新基因LX3在细胞内的表达定位情况。结果获得真核表达载体pEGFP-N1/LX3,并成功转染293T和NIH3T3细胞,共聚焦荧光显微镜观察了LX3-GFP在细胞内的荧光分布。结论IL-6诱导相关新基因在细胞内为全细胞分布。Objective To analyze the expressional localization of novel gene LX3 induced by IL-6. Methods The LX3 eDNA was cloned into the eukaryotie expression vector pEGFP-N1, which containing green fluorescence protein (GFP) gene, and then the recombinant was transfected into 293T cell and NIH3T3 cell by Lipofection. The LX3-GFP fusion protein was detected by laser eonfocal microscopy. Results The eukaryotic expression vector pEGFP-N1-LX3 was constructed successfully; the LX3 gene was transfected into 293T cell and NIH3T3 cell by Lipofection. The location of LX3 expression in cultivated cells was detected by laser confocal microscopy. Conclusion LX3 gene proves to be expressed in the whole cell.
