摘 要:目的观察大鼠去卵巢后骨髓细胞核因子κB受体活化子配体(RANKL)、核因子κB受体活化子(RANK)、护骨素(OPG)以及TNFα基因表达的改变和雌激素的影响。方法健康3m龄雌性SD大鼠72只,24只为假手术对照组,48只去卵巢,随机分为去卵巢组和雌激素组,每组24只。分别于去卵巢后2、4、6、12w每组各取6只大鼠骨髓细胞提取RNA,RTPCR半定量分析其表达。结果与对照组相比,去卵巢后2w,去卵巢组骨髓细胞TNFα和RANKLmRNA表达显著升高(P<0.05~0.01),第4~6w,上述改变达高峰,至第12wTNFα仍呈有意义增高;而OPG表达在第2~6w则明显降低(P<0.01),2~4w为最低点;OPGRANKL比值2~6w为最低(P<0.01),12w仍低于对照组。雌激素组以上各时间TNFα和RANKL表达低于去卵巢组(P<0.05,P<0.01)而OPG表达和OPGRANKL比值明显高于去卵巢组(P<0.01)。各组全程未见RANK基因表达水平有明显变化。结论雌激素抑制大鼠去卵巢后骨髓细胞过度表达RANKL和TNFα而增加OPG的表达。Objective To observe the changes of gene expression of receptor activator of NF-κB ligand( RANKL), receptor activator of NF-κB (RANK), osteoprotegerin (OPG) and tumor necrosis faetor-α(TNF-α) in bone marrow of ovariectomyized rats and the effects of estrogen treatment. Methods Seventy-two healthy female SD rats,3 months old, were randomly divided into three groups(24 in each) : the sham-operated control group( A), the ovariectomized group(B) and the estrogen treated group after ovafiectomy (C). Bone marrow cells of 6 rats from each group were respectively collected at four different time points (2nd ,4^th ,6^th and 12^th weeks post operation) and total RNA was extracted by TRIZOL.Resuits From the 2nd week after the operation, RANKL and TNF-α genes expression was significantly increased in Group B( P 〈 0.05 or P 〈 0.01 ) and reached the peak in the 4^th to 6^th week. The increased expression of TNF-α was maintained till the 12^th week. In the 2^nd to 6^th week, OPG gene expression decreased significantly in group B. The lowest level were at 2^nd to 4^th week. The OPG/RANKL ratio reached the lowest level in 2^nd to 6^th week till 12^th week. No significant change of RANK gene expression was revealed in the whole courses. Conclusions Estrogen inhibits the over expression of TNF-α and RANKL gene and promote OPG gene expression in bone marrow of rats after ovariectomy.
