摘 要:取翘嘴鲌的前、中、后肠和肝胰脏,分别用福林-酚法、淀粉-碘显色法和聚乙烯醇橄榄油乳化液水解法测定了肠道和肝胰脏的蛋白酶、淀粉酶和脂肪酶比活力,同时研究了不同温度(20,25,30,35,40,45,50,55℃)、不同pH值(5.6,6.0,6.4,6.8,7.0,7.2,7.6,8.0)对其消化酶比活力的影响.结果表明:翘嘴鲌肠道消化酶比活力显著高于肝胰脏(P<0.05).前肠、中肠和后肠的蛋白酶、淀粉酶、脂肪酶的比活力差异不显著(P>0.05).前肠、中肠、后肠及肝胰脏的蛋白酶适宜的pH值分别为6.8~7.2,6.8~7.2,6.8~7.0,6.8~7.0;淀粉酶适宜的pH值分别为6.4~7.6,7.0~7.2,7.0~7.2,6.0~7.0;脂肪酶适宜的pH值分别为7.0~7.6,6.8~7.2,6.8~7.6,7.2.前肠、中肠、后肠及肝胰脏的蛋白酶适宜的温度分别为35~45,35~45,35~45,30~40℃;淀粉酶适宜的温度分别为30~40,35~40,40~45,30~45℃;脂肪酶适宜的温度分别为40~45,35~45,30~50,35℃.Protease, amyase and lipase specific activity in the intestine and hepatopanereas was measured respectively through folin-phenol method, the way of starch-iodine reaction and turbidimetric method to examine the digestive enzyme activity of Culter alburnus juveniles. Meanwhile, digestive enzyme activity in fore, middle and hind intestine and hepatopancreas was measured to study the effect of temperature (20,25,30,35,40,45,50,55℃) and pH (5.6,6.0,6.4,6.8,7.0,7.2,7.6,8.0) on digestive enzyme activity . The results showed that activity of digestive enzymes in the intestine of Culter a/burnus was significantly higher than that of hepatopancreas( P 〈 0.05). There was no significant difference of protease and amyase and lipase activity from fore-gut to hind-gut of Culter alburnus ( P 〉 0. 05). The optimum pH value of protease in fore-gut and middle-gut and hind-gut and hepatopancreas is 6.8 - 7.2,6.8 - 7.2, 6.8 - 7.0,6.8 - 7.0 , that of amyase is 6.4 - 7.6,7.0 - 7.2,7.0 - 7.2,6.0 - 7.0, that of lipase is 7.0 - 7.6,6.8 - 7.2,6.8 - 7.6,7.2 respectively. The optimum temperature of protease in fore-gut and middle-gut and hind-gut and hepatopancreas is 35 - 45,35 - 45,35 - 45,30 - 40℃, that of amyase is 30 - 40,35 - 40,40 - 45,30 - 45 ℃, that of lipase is 40 - 45,35 - 45,30 -50 and 35℃ respectively. This study provides the basic information for digestive physiology of Culter alburnus.
