摘  要:目的 研究复方黄芪提取物(CAE)对细菌脂多糖诱导大鼠腹腔巨噬细胞(PMφS)释放肿瘤坏死因子((TNFα)、白细胞介素-1(IL-1)及一氧化氮(NO)的影响。方法 10mg/L LPS体外诱导大鼠PMφS分泌TNFα、IL-1与NO等因子,采用小鼠成纤维细胞瘤株(L929)杀伤法检测TNFα的含量、小鼠胸腺细胞增殖法检测IL-1的活性、Griess法检测NO的水平。结果 CAE(11.3-90mg·L^-1)能显著抑制LPS激活的巨噬细胞对TNFα、IL-1与NO的分泌。结论 CAE抑制巨噬细胞的分泌功能可能是其保肝作用的机制之一。Aim To study effect of compound astragalus extract (CAE)on TNFα, IL-1 and NO secreted by normal rat peritoneal macrophages (PMφ). Methods Rat peritoneal macrophages was stimulated with 10 mg · L^-1 LPS in vitro. TNFα was measured by assay of cytotoxicity against L929 cells ,The concentration of NO was determined by Griess assay, activity of IL-1 was assayed by [ ^3H ] TdR incorporation assay. Results CAE could markedly inhibit production of TNFα, IL-1 and NO from normal rats PMφS induced by LPS. Conclusion Inhibiting directly secretion of TNFα, IL-1 and NO produced by PMφS might be one of the mechanisms of hepatoproteetive effect of CAE.
