摘 要:目的观察饱和脂肪酸对大鼠主动脉MMP-9及PPARα表达的影响及非诺贝特对其的保护作用。方法雄性Wistar大鼠30只,随机分为对照组、高脂组(HF组)、高脂加非诺贝特组(HF+FF组)。分组喂养20周后,采用RT-PCR方法测定大鼠主动脉MMP-9和PPARα的mRNA水平;采用Western blot方法测定大鼠主动脉MMP-9和PPARα蛋白表达水平。结果HF组大鼠主动脉MMP-9 mRNA及其蛋白水平均明显高于对照组(t=4.65、5.48,P<0.01);PPARαmRNA及其蛋白水平则低于对照组(t=2.52、2.98,P<0.05)。HF+FF组PPARαmRNA及其蛋白水平明显高于HF组(t=4.51、6.29,P<0.01),而MMP-9 mRNA及其蛋白水平则明显低于HF组(t=5.62、5.97,P<0.01)。结论非诺贝特可上调PPARα表达,下调MMP-9水平,从而对动脉粥样硬化的发生和发展可能起到保护作用。Objective To study the effect of high fat diet on the expression of MMP 9 and PPARα in the rat aorta and the protective effect of Fenofibrate. Methods Thirty male Wistar rats were randomly divided into control group, high fat diet group (HF) and Fenofibrate treated group (HF + FF). Fenofibrate was administrated orally for 20 weeks, plasma triglyceride and cholesterol levels were measured. Protein levels and mRNA levels of MMP-9 and PPARα of rat aorta were determined using Western blot and RT-PCR. Results In HF group, both the mRNA and its protein levels of MMP 9 in rat aorta were higher than that in the control group (t= 4.65,5.48; P〈0. 05), while the expression of PPARaand its protein were lower (t=2.52,2.98; P〈 0.05). In HF+FF group, the expression of PPARα mRNA and its protein increased compared with HF group (t= 4.51,6.29, P〈0.01), while MMP-9 mRNA and its protein decreased (t=5.62,5.97. P〈0.01). Conclusion Fenofibrate may up-regulate the expression of PPARα and down-regulate that of MMP-9, thus to play a role in the protection of the occurrence and development of atherosclerosis.
