摘  要:目的研究IPF1基因启动子区G3/G4多态性位点与鲁北地区汉族人群糖尿病的相关性。方法选择2型糖尿病96例、1型糖尿病36例及正常对照组100例作为研究对象,对IPF1基因启动子区G3/G4多态性位点进行PCR扩增,PCR产物纯化后,经ABI PRISM 3700 DNA全自动测序仪直接测序,然后进行基因分型,并通过相关分析,研究该位点与糖尿病的相关性。结果对照组与1型、2型糖尿病组的基因型频率和等位基因频率分布差异无统计学意义(χ2=8.566,P=0.073,χ2=5.944,P=0.051)。结论IPF1基因启动子区G3/G4多态性位点与中国汉族人群糖尿病无显著相关性,暂不能作为预测中国汉族人发生糖尿病危险性的遗传学标志。Objective To study the association of single nucleotide polymorphism (G3/G4) in the promoter region of insulin promoter factor- 1 gene with diabetes of Han nationality in north Shandong. Methods In this study, 96 type 2 diabetic patients, 36 type 1 diabetic patients and 100 normal control subjects were enrolled. G3/G4 polymorphism in the promoter region of insulin promoter factor - 1 gene was amplified by PCR. Then, PCR products were purified and sequenced by ABI PRISM 3700 DNA sequencer. Subsequently, the SNP was genotyped and, its association with diabetes was examined. Results There was no significant difference of genotype frequency and allele frequency between control group and type 1 or 2 diabetic group(χ^2 = 8. 566, P = 0. 073; χ^2 = 5. 944, P = 0. 051 ). Conclusion G3/G4 polymorphism in the promoter region of insulin promoter factor- 1 gene is not markedly related to diabetes and may not be used as a genetic marker in predicting the risk of developing DM in Chinese Han nationality.
