摘 要:目的:探讨重症急性胰腺炎在早期时肺组织中MDA、MPO的变化与意义。方法:健康成年雄性SD大鼠40只,按随机化原则分成5组,每组8只大鼠。即假手术组、重症急性胰腺炎30、60、120和360min组(SAP组:胰胆管逆行注入5%牛磺胆酸钠溶液),剖杀各组大鼠,分别检测肺组织中丙二醛含量(MDA)和髓过氧化物酶(MPO)活性。结果:假手术组动物肺组织MDA为(1.61±0.62)nmol/mg,MPO为(2.63±0.74)U/g。重症急性胰腺炎模型制成后30、60、120和360min大鼠肺组织MDA和MPO均升高,分别为(1.91±0.94)nmol/mg、(2.17±0.64)nmol/mg、(2.54±1.14)nmol/mg、(4.99±1.68)nmol/mg和(3.03±0.83)U/g、(3.88±1.08)U/g、(4.57±1.16)U/g、(6.43±1.15)U/g。在60min时MPO明显升高(P<0.05);在360min时MDA显著升高(P<0.01)。结论:试验性大鼠重症急性胰腺炎早期(360min)时,肺组织中出现脂质过氧化反应和大量中性粒细胞浸润。Objective :To investigate the changes and signifieanee of pulmonary MDA and MPO induced by severe acute pancreatitis during its early stage. Methods : Forty adult male Sprague-Dawley rats weighting 300-400g were randomly divided into five groups with 8 ones in each sham-operation group, SAP 30,60,120 and 360-minute group. The models of SAP were induced by retrograde injection of 5 % sodium taurocholate into the biliopancreatic duct of rats. MDA levels and MPO activity in pulmonary tissue were measured by dissecting each rat at a different time interval. Results : Pulmonary MDA and MPO levels in the sham group were (1.61± 0.62)nmol/mg and (2.63 ± 0.74) U/g, respectively. After the model of SAP were induced, pulmonary MDA and MPO levels increased at the phase of 30,60,120 and 360 minute. They were(1.91 ± 0.94) ritual/rag, (2.17 ± 0.64) nmol/mg, (2.54 ± 1.14) nmol/mg, (4.99 ± 1.68) nmol/mg and ( 3.03 ± 0.83 ) U/g, ( 3.88 ± 1.08)U/g,(4.57± 1.16)U/g,(6.43 ± 1.15)U/g, respectively. MPO levels were increased significantly at 60 minute (P 〈 0.05) while MDA levels were seen significant increase at 360 minute( P 〈 0.01 ). Conclusion :Lipid peroxidation and much PMN infiltration exist in pal- monary tissue during SAP early stage in rats.
