摘  要:目的探讨康莱特与阿霉素联合应用的体外抗肿瘤活性及其相关机制。方法将康莱特与阿霉素单独及联合应用于骨肉瘤细胞,采用MTT法检测细胞毒性作用,流式细胞仪定量分析凋亡细胞所占比例,倒置相差显微镜、荧光显微镜下观察凋亡细胞形态改变。结果康莱特质量浓度达到1 g.L-1时,细胞抑制率不再明显改变,但如与1 mg.L-1阿霉素合用将使细胞抑制率进一步提升(P<0.01)。细胞形态观察及凋亡率测定也显示,康莱特与阿霉素联用可诱导更多骨肉瘤细胞凋亡。结论康莱特与阿霉素合用可高效杀伤肿瘤细胞。Objective To study the antitumor activity in vitro of Kanglaite combined with doxorubicin and related mechanism. Methods Kanglaite and doxorubicin were applied on the OS-732 cells respectively or jointly and inhibition rate was measured by MTT assay; The celluar shape were observed with inverted phase contrast microscope and fluorescence microscope; The apoptotic rate was analyzed with flow cytometry (FCM). Results Kanglaite had the effect of up-regulating Fas expression and inducing apoptosls; the inhibition rate was stagnated with 1 g· L^-1 Kanglaite, if combined with 1 mg· L^-1 doxorubicin, the inhibition rate was increased obviously(P 〈 0.01 ). Changes of Celluar shape and apoptotic rate also showed apoptosisinducing effect of joint application , which was much stronger than those of respective application. Conclusions Combination of Kanglaite and doxorubicin show a strong killing effect on OS-732 cells.
