摘  要:目的观察束缚应激对局灶性脑缺血大鼠的运动行为及脑内促肾上腺皮质激素释放因子(CRF)表达的影响。方法雄性SD大鼠40只,随机分为假手术组和缺血组,每组20只,采用线栓法制作脑缺血再灌注(tMCAO)模型。术后第28天再将每组大鼠随机分为单纯假手术组、假手术+应激组、单纯tMCAO组和tMCAO+应激组。从手术处理后第5周开始行2周束缚应激,应激结束后行抑郁行为测试。用免疫组化方法检测大鼠脑内CRF的表达情况,观察束缚应激对局灶性脑缺血大鼠脑内CRF表达的影响。结果①束缚应激使局灶性脑缺血大鼠的水平运动和垂直运动得分均显著减少,蔗糖水偏好比明显降低,与单纯假手术组、假手术+应激组及单纯tMCAO组相比,差异均有显著性(P<0.01);②tMCAO+应激组大鼠脑内下丘脑室旁核、杏仁核中央亚核、终纹床核中CRF阳性神经元数量与其他组比较明显增多,差异有显著性(P<0.01)。结论施加束缚应激后,局灶性脑缺血大鼠出现明显的抑郁行为,脑内参与调节情感的核团内CRF蛋白含量增加,表达反应性增强,考虑可能是卒中后抑郁的生物学机制之一。Objective To observe the effect of restraint stress on behavior and corticotropin-releasing factor (CRF) expression after focal cerebral ischemia in rats. Methods Forty male Sprague-Dawley rats were randomly assigned to sham-operation group and ischemia-operation group. A rat model of cerebral ischemia-reperfusion (transient middle cerebral artery occlusion, tMCAO) was achieved by the suture method. On day 28 after surgery, the animals in beth groups were randomly allocated into sham-operation group only and sham-operation + stress group, the tMCAO group only and tMCAO + stress group. Two-week restraint stress was performed from the fifth week after surgery intervention. Depressive behavior was tested after the restraint stress. CRF expression in rat brains was detected by immunohistocbemistry method, and the effects of restraint stress on CRF expression in focal cerebral ischemic rats were observed. Results Restraint stress significantly reduced the scores of beth vertical and horizontal motions in rats with focal cerebral ischemia, and their preference for the sucrose solution significantly reduced compared with the shamoperation group only, the sham-operation + stress group and the tMCAO group only, there were significant differences among the groups (P 〈 0. 01 ). The numbers of CRF positive neuron increased significantly in the paraventricular hypothalamic nucleus, the central nucleus of the amygdala and the bed nucleus of stria terminalis in the tMCAO + stress group compared with other groups, and there were statistical significant differences among the groups ( P 〈 0. 01 ). Conclusion After restraint stress was performed, the focal cerebral ischemic rats developed significant depressive behavior. CRF protein content increased within the nuclei that involved in adjusting emotion in brains, and its expressive reactivity increased. It may be one of the biological mechanisms of poststroke depression.
