摘 要:目的:建立测定小鼠血浆中蒜氨酸浓度的液相色谱-质谱法及其在小鼠体内的药动学研究。方法:使用大气压化学电离源(APCI),在正离子条件下选择性离子监测(SIM)。用于定量分析的离子分别为[M+H]^+178→[M+Na]^+200(蒜氨酸)和[M+H]^+195→[M+Na]^+217(内标,咖啡因)。结果:小鼠灌胃给蒜氨酸100 mg·kg^(-1)后,其主要的药动学参数为:CL=0.01 mg·min^(-1)·kg^(-1),K_(12)=0.057 min^(-1),K_(21)=0.024 min^(-1),K_a=0.127 min^(-1),t_(1/2α)=6.89 min^(-1),t_(1/2β)=112.34 min^(-1),AUC=5046.27μg·min^(-1)·mL^(-1)。结论:此法灵敏、快速、准确,可用于小鼠血浆中蒜氨酸的浓度测定及临床前药动学研究。Objective: To develop HPLC - MS method for determination of alliin concentration in mice plasma and in vivo pharmacokinetics study. Methods: The plasma samples were extracted with methanol. HP - MSD equipped with an atmospheric pressure chemical ionization(APCI) source was used as detector and was operated in positive ion mode. Selected ionization monitoring was used and transitions selected for quantitation were : [ M + H ]^+ 178→ [ M + Na]^+ 200 for alliin and[ M + H]^+ 195→[ M + Na]^ + 217 for caffeine. Results: After single dose of 100 mg ~ kg- ~ in mice, the main pharmacokinetic parameters were estimated to be as follows: CL=0.01mg·min^-1·kg^-1,K12=0.057min^-1,K21=0.024min^-1,K=0.127min6-1,t1/2a=6.89min^-1,t1/2β=112.34min^-1,AUC=5046.27μg·min^-1·mL^-1 Conclusion: The method is sensitive, simple and accurate, and will be suitable in determination of alliin in plasma and its pharmacodynamics.
