摘 要:目的观察外源性给予硫化氢(H2S)对血管钙化的影响。方法用肌注维生素D3和灌胃尼古丁诱导大鼠血管钙化,运用Von Kossa染色、原子吸收测定钙含量4、5Ca2+沉积及碱性磷酸酶(ALP)活性测定判断血管钙化程度,半定量RT-PCR方法测定骨桥蛋白(OPN)mRNA水平。结果钙化组大鼠血管Von Kossa染色见平滑肌细胞内和细胞间基质有大量黑色颗粒聚集;主动脉钙含量、45Ca2+沉积及ALP活性分别较对照高6.8倍、1.4倍和1.9倍(P<0.01),OPN mRNA表达明显上调;低、高剂量NaHS组均明显缓解上述指标的变化(P<0.01)。结论补充H2S可减轻钙化血管的钙化程度。objective To observe the effect of exogenous H2S on vascular calcification in rat. Methods vascular calcification in rat was induced by administration of Vitamin D3 plus nicotine. To Estimate the extent of calcification by Von Kossa staining, calcium content, ^45Ca^2+ deposition and alkaline phosphatase activity. Osteopontin (OPN) mRNA was determined by competitive quantitative RT-PCR. Results A strong positive staining of black/brown are- as among the elastic fibers of the medial layer in calcified aorta by Von Kossa staining, Calcium content, ^45Ca^2+ accumulation and ALP activity in calcified arteries increased by 6.77 -, 1.42 -, and 1.87 - fold as compared with the controls ;The expression of OPN mRNA is up-regulated (P 〈 0. 01 ). However, calcium content, ^45Ca^2+ uptake and ALP activity in VDN plus Naris group were lower than those in VDN group. Low and high dosage Naris obviously relieve degree of calcification in the cardiovascular tissues in a dosage-dependent manner (P 〈 0. 01 ). Conclusion H2S plays a role in the pathogenesis of vascular calcification by reducing vascular calcification.
