摘  要:目的:探讨2-甲氧雌二醇(2-ME)对人子宫内膜癌细胞株KLE细胞凋亡作用的影响。方法:将体外培养的人子宫内膜癌细胞株KLE细胞分为实验组和对照组,其中实验组培养液中加入不同浓度的2-ME,对照组不含2-ME。通过流式细胞仪(FCM)观察KLE细胞的凋亡率及凋亡基因p53和Bcl-2在诱导细胞凋亡中的作用。结果:实验组2-ME浓度为5.0μM-20.0μM时,出现细胞凋亡率升高,与对照组相比有显著性差异(P<0.05),实验组2-ME浓度达2.0μM以上时p53和Bcl-2阳性表达率与对照组相比明显下降(P<0.05)。结论:2-ME对体外培养的人子宫内膜癌KLE细胞株有促进凋亡作用。Objective: To investigate the effect of 2-methoxyestradiol(2-ME) on apoptosis in human endometrial carcinoma cell line( KLE cells) in vitro. Methods: Endometrial carcinoma cell line( KLE cells) were divided into study group( coltured with 2- ME in different concentrations) and control group (c oltured without 2-ME). Apoptotic rate and expression of tumor suppressor gene p53 and Bcl-2 were detected by flow cytometry(FCM). Results: The apoptotic rates in study group with 2-ME concentrations between 5 ~20μ M were significantly higher than that in control group(P 〈0. 05 ). The positive expression rate of p53 and Bcl-2 in study group with the 2-ME beyond 2μM were significantly lower than those in control group (P 〈 0.05 ). Conclusion: The study suggests that 2-ME can induce the apoptosis of KLE ceils in vitro.
