摘 要:目的探讨东北汉族人群WNK4基因多态性与原发性高血压的关系。方法259例原发性高血压患者和235例健康正常对照人群血样应用PCR-RFLP方法检测A589S多态,结果经测序验证,同时检测所有人群的血糖、血脂等生化指标。结果(1)两组研究对象基因型频率间差别无显著性意义(P>0.05),但等位基因频率间差别有显著性意义(P<0.05);(2)GT和TT基因型与收缩压及舒张压升高密切相关(P=0.041,0.032)。结论WNK4基因A589S多态可能与高血压密切相关。Objective To elucidate the association of WNK4 gene A589S polymorphism with essential hypertension in north - eastern han isolated population. Methods Genotyping was performed using polymerase chain reaction - restriction fragment length polymorphism (PCR- RFLP) and confirmed by direct sequencing. 259 unrelated patients with essential hypertension and 235 unrelated normal controls were recruited. All the subjects had routine laboratory tests. Results ( 1 ) The frequency of WNK4 genotype GG, GT and TT were 0. 63, 0. 33, 0. 04 in controls respectively, and 0. 53, 0. 43, 0. 04 in hypertensive respectively. There was no significant differences in genotypes between hypertensive group and normal controls. ( P 〉 0. 05 ). The frequencies of G and T were 0. 80, 0. 20 in normal controls and 0. 74, 0. 26 in hypertensives respectively. And the frequencies of T allele in hypertension group is significantly higher than that in controls (25.9% versus 20. 2%, P 〈 0. 05 ). (2) The GT and TT genotypes were closely associated with raised systolic and diastolic blood pressure ( P =0. 041,0. 032). Conclusion The WNK4 gene A589S polymorphism is associated with essential hypertension in north -eastern han population.
