摘 要:目的:研究难治性癫患者脑组织内钙黏蛋白-18(cadherin-18)mRNA的表达,拟从另一角度加深对人类难治性癫的认识。方法:在基因芯片扫描的基础上,运用RT-PCR检测钙黏蛋白-18mRNA在难治性癫患者脑部的变化。结果:基因芯片扫描显示钙黏蛋白-18mRNA在难治性癫患者脑部比正常人表达上调5.320倍,RT-PCR检测同样显示钙黏蛋白-18mRNA在难治性癫患者脑部表达增强,其电泳条带灰度值为25.547,而正常组仅为11.176。结论:结果表明特异性表达于中枢神经系统的钙黏蛋白-18mRNA在难治性癫患者脑部表达增强,提示钙黏蛋白-18可能参与了人类难治性癫的形成。Aim: To study the change of cadherin-18 mRNA expression in the brain of intractable epileptic patients so as to explore the possible mechanisms of the pathogenesis of intractable epilepsy. Methods: The microarray scanning and reverse transcription polymerase chain reaction (RT-PCR) were used to evaluate cadherin-18 mRNA expression in the brain of 40 intractable epileptic patients and 8 normal control human. Results: As compared with the normal control group, there appeared a significant increase in cadherin-18 mRNA expression in intractable epileptic patients, with microarray scanning cy5/cy3 ratio being 5.320, RT- PCR gray density of electrophoresis band being 25.547 in intractable epileptic patient group, 11.176 in normal control group, respectively. Conclusion: It is cadherin-18 mRNA, not N-cadherin or any other cadherin mRNA found to have different expression in intractable epilepsy of human. Cadherin-18 may play an important role in the pathogenesis of intractable epilepsy of human.
