摘  要:目的:研究髓系白血病细胞系K562和HL60分化抗原的表达,分析其免疫学表型特征。方法:收集RP MI1640培养3d的对数生长期K562和HL60细胞,用流式细胞仪测定细胞膜表面CD7、CD11b、CD13、CD14、CD33、CD34、CD38、CD64、CD41a、CD117和HLA DR11种分化抗原表达百分率,了解其免疫学表型,同时进行瑞特姬姆萨(Wright Giemsa)染色观察细胞形态和髓过氧化物酶染色(MPO)。结果:K562白血病细胞的髓系特征分化抗原CD13和CD117表达率较低,为5.3%和5.2%,其他分化抗原CD11b、CD14、CD33、CD64、CD41a和反映细胞阶段性特征的分化抗原CD7、CD34、CD38和HLA DR表达均处于极低水平,MPO染色为阴性;HL60白血病细胞的髓系特征分化抗原CD13表达率高达94.0%,仅伴随有少量CD11b、CD64和CD41a表达,祖细胞特征的CD38表达率为36.7%,MPO染色为阳性;细胞形态学上K562细胞原始未分化特性比较明显,HL60细胞质内已有嗜天青颗粒和空泡等细胞分化特征。结论:K562细胞的免疫学表型和形态学均表现出未分化细胞特征,因其具有多项分化潜能,因此更适合进行细胞多向分化研究。HL60的髓系祖细胞特性明显,适合进行粒系、单核系分化研究。Aim: To study differentiation antigens and immunology phenotype characteristics of the myeloid leukemia cell-lines of K562 and HL60. Methods: K562 and HL60 cells cultured for 3 d by PRMI1640 in logarithm growth period were collected, the expression percentage of the differentiation antigens located at the cell membrane surface was detected by flow cytometer (FCM) , and at the same time the morphology of cells were observed by staining with Wright-Giemsa dyes. Results: The expression rates of myeloid differentiation antigens CD13 and CD117 were relatively low in K562 cells (5.3% and 5.2% respectively). The expressing rates of other myeloid differentiation antigens including CD11b, CD14, CD33, CD64, CD41a and the cell stage differentiation antigens including CD7, CD34, CD38 and HLA-DR were in a very low level;MPO was negative in K562 cells. In HL60 cells, the expressing rate of CD13 was as high as 94.0% , and expression rates of CD11b, CD64 and CD41a were lower. CD38 which reflected progenitor cell characteristic had a high expression rate of 36.7% in HL60 cells. MPO was positive in HL60 cells. K562 cells morphology obviously showed a primitive and no-differentiation characteristic. The cytoplasm of HL60 cells was observed the mauve granules and vacuoles, which indicated the differentiation of HL60 cells. Conclusion: The immunology phenotype and morphology of K562 cells demon strate no-differentiation cell characteristies,K562 cells have multiple linage differentiation abitity and are more suitable for the study on multiple orientation differentiation of cells ,while HL60 cells have obvious characteristies of mycloid progenitor cell and therefore are suitable for the study on granulocyte or monocyte differentiation.
