摘　　要：【目的】探讨选择性环氧化酶-2(COX-2)抑制剂Celebrex对膀胱癌T24细胞株COX-2 mRNA和前列腺素E2(PGE2)表达的影响。【方法】体外培养人膀胱癌细胞株T24细胞,应用逆转录聚合酶链反应(RT-PCR)和酶联免疫吸附测定(ELISA)方法,检测选择性COX-2抑制剂Celebrex与膀胱癌T24细胞株COX-2 mRNA和PGE2表达的时间-效应关系和剂量-效应关系。【结果】Celebrex在一定时间和剂量范围内可明显抑制膀胱癌T24细胞株PGE2的表达,但Celebrex无论在时间-效应或剂量-效应关系上均不影响人膀胱癌T24细胞株COX-2 mRNA的表达。【结论】Celebrex对COX-2 mRNA的表达无明显影响,主要是通过抑制COX-2酶及其主要代谢产物PGE2的表达而发挥抗肿瘤效应。[Objective]To study the effects of the selective eyelooxygenase-2（COX-2） inhibitor ,eelebrex, on the expression of COX-2 mRNA and PGE2 in bladder cancer T24 cell lines. [Methods]The time-effect relationship and concentration-effect relationship between eelebrex and the expression of COX-2 mRNA and PGFa in bladder cancer T24 cell lines were studied by enzyme-linked immuno-absordent assay（ELISA） and reverse transeriptase polymerase chain reaetion（RT-PCR）. [Results]The expression of PGE2 was inhibited by eelebrex in a certain degree of time-dependent and dose-dependent manner, while the expression of COX-2 mRNA was not inhibited by eelebrex. [Conclusion]The expression of COX-2 mRNA is not inhibited by eelebrex. Inhibitions of the the expression of COX-2 protein and PGE2 in bladder cancer T24 cell lines are the main mechanisms by which eelebrex suppresses cancers.
