摘 要:目的:探讨小鼠角膜缝线及联合结膜下药物注射动物模型B7-1 mRNA表达与非特异性角膜炎症反应的相关性及核因子NF-kB抑制剂对B7-1 mRNA表达的阻断作用。方法:建立BALB/C鼠角膜缝线及联合结膜下药物注射的动物模型,通过逆转录聚合酶链反应(RT-PCR)对各组术后1、3、7和14 d不同时相点角膜B7-1 mRNA的转录水平进行检测。结果:缝线联合细菌脂多糖(LPS)1、3和7 d角膜处理组与单纯缝线组比较B7-1 mRNA表达增加(P<0.05),而术后14 d两组比较差异无显著性(P>0.05);缝线联合LPS和吡咯啉烷二硫代氨基甲酸酯(PDTC)角膜处理组1、7和14 d与缝线联合LPS组比较B7-1 mRNA表达降低,3 d组B7-1 mRNA表达增加(P<0.05)。结论:缝线联合结膜下LPS注射可在术后诱导B7-1 mRNA的高水平表达,使炎症反应程度加重;而核因子NF-kB抑制剂PDTC可抑制角膜B7-1 mRNA的表达。核因子NF-kB对B7-1 mRNA表达具有重要的调控作用。Objective To establish the mouse nonspecific keratitis model by corneal suture combined with subconjunctival injection in order to analyze the relationship between B7-1 mRNA expression and corneal inflammation and discuss the inhibition of nuclear factor-kB (NF-kB) inhibitor on B7-1 mRNA expression. Methods The corneal suture or combined with subconjunctival injection models were constructed in BALB/C mice. The corneas of each group were excised at the 1st, 2nd, 7th, 14th day postoperatively and the B7-1 mRNA expressions were investigated by reverse transcription polymerase chain reaction (RT-PCR). Results At 1st, 3rd and 7th day postoperatively the expression of corneal B7-1 mRNA increased by the inducement of corneal suture combined with lipopolysaccharide (LPS) subconjunctival injection than that by the ,corneal suture (P〈0. 05). At 14th day postoperatively there was no significant difference of the expression of corneal B7 1 mRNA between two groups (P〉0.05); At 1st, 7th, 14th day postoperatively the expression of corneal B7-1 mRNA decreased, in the group of inducement of corneal suture combined with LPS and pyrrolidine dithiocarbamate (PDTC) subconjunctival injection compared with the group of inducement of corneal suture combined with LPS subconjunctival injection (P〈0.05), but it increased at 3rd day postoperatively. Conclusion A high expression of B7-1 mRNA can be induced by the corneal stuture combined with LPS subconjunctival injection, which has a close relation with the deteriorated inflammation. The inhibitor (PDTC) of NF-kB can inhibit the expression of B7-1 mRNA. The activation of NF-kB has a great influence in the regulation of BT-1 mRNA expression.
