摘  要:目的探讨兔尸体肺组织在不同灌注条件下、不同时间点,一氧化氮合酶(NOS)同功酶表达变化的规律。方法48只健康日本大耳白兔随机分成6组:正常组(A组)、单纯给药组(B组)、LPD液灌注组(C组)、EC液灌注组(D组)、给药后LPD液灌注组(E组)、给药后EC液灌注组(F组),每组8只。建立尸体肺灌注模型,采用免疫组织化学SABC法及RT-PCR技术检测不同时间点再灌注肺组织内皮型(eNOS)和诱导型(iNOS)一氧化氮合酶及mRNA表达的变化。结果NOS的免疫组化结果如下:①A、B组iNOS明显表达,eNOS表达明显下调。②C、D组iNOS不同程度表达,eNOS表达下调。③E、F组iNOS轻度表达,eNOS表达轻度下调。eNOS、iNOSmRNA的表达变化趋势与免疫组化结果一致。结论尸体肺组织eNOS、iNOS的表达,与肺组织缺血时间、再灌注条件密切相关。Objective To investigate the experssion of nitric oxide synthase(NOS) on isolated corpse lung in rabbit model under different perfusion condition. Methods 48 rabbits were randomly divided into 6 groups : normal (group A) ;medication administration only (group B) ; perfused with fluid LPD (group C) ; perfused with fluid EC (group D) ; perfused with fluid LPD after medication administration (group E) and perfused with fluid EC after medication administration (gronp F), each group 8 rabbits. Perfusion model of isolated lungs were established, lmmunohistochemical technique and RT- PCR method were used to determine the various protein and nlRNA expression of eNOS, iNOS in reperfusion pulmonary tissue at different time point. Results The expressions of NOS were as follows: (1) Significant expression of iNOS was shown in group A and B, but expression of eNOS was lowed much in endothelial cells;(2) iNOS expressions were variable in group C and D, and that of eNOS was decreased;(3)iNOS expressions in group E and F were imponderability, and eNOS decreased slightly. Similar result were revealed by RT - PCR method. Conclusion The decrease of eNOS and increase of iNOS have close correlation with ischemia time and reperfusion condition on isolated corpse lung.
