摘 要:目的:观察灵芝对大鼠退变颈椎间盘中促进椎间盘炎症发生的重要介质白细胞介素1β、肿瘤坏死因子α、一氧化氮的含量的影响,并分析其意义。方法:实验于2002-12/2003-07在复旦大学医学院实验动物中心完成。选择6月龄雌性SD大鼠24只,随机数字表法分为正常组、模型组和灵芝组,每组8只。模型组和灵芝组大鼠通过手术建立动静力失衡性颈椎病模型,诱导大鼠颈椎间盘退变。术后4个月开始灌胃,灵芝组大鼠予400mg/kg剂量灵芝穴由上海绿谷集团提供的灵芝孢子粉雪,以10mL饮用水配制成混悬液灌胃,其余两组予同等体积的饮用水,1次/d,共2个月。然后取大鼠C3-4、C4-5、C5-6椎间盘标本进行组织形态学观察,同时应用ELASA法检测白细胞介素1β、肿瘤坏死因子α的含量,应用吸光度法检测一氧化氮的含量,并进行组间比较。结果:实验动物24只全部进入结果分析,没有脱落。①实验6个月后大鼠颈椎间盘显微镜下的组织形态学观察:正常组大鼠见椎间盘纤维环出现裂隙,排列轻度不规则,髓核出现皱缩变小,软骨板可见不规则增生;模型组大鼠见不到椎间盘的正常组织,代之以肥大的软骨细胞;灵芝组大鼠椎间盘髓核完全纤维化,纤维环层状结构不明显。②与正常组比较,模型组大鼠颈椎间盘内白细胞介素1β、肿瘤坏死因子α和一氧化氮的含量显著增高(P<0.01)。③灵芝组大鼠颈椎间盘内白细胞介素1β、肿瘤坏死因子α和一氧化氮的含量较模型组低,差异有显著性意义穴P<0.05雪。结论:退变颈椎间盘中释放的白细胞介素1β、肿瘤坏死因子α和一氧化氮可加速椎间盘的退变,中药灵芝可抑制这些因子的释放,可能具有延缓椎间盘退变的作用。AIM: To observe the effects of Lingzhi on contents of interleukin-1 (IL)-1β, tumor necrosis factor (TNF)-α and nitric oxide (NO) in degenerated cervical intervertebral disc, which can accelerate the inflammation of intervertebral disc, and analyze the significances. METHODS: The experiment was conducted in the Experimental Animal Center, Medical Collage of Fudan University from December 2002 to July 2003. A total of 24 six-month-old female SD rats were randomly divided into control group, model group and Lingzhi group with 8 rats in each group. Rats in the model group and lingzhi group were made into dynamic and static force-imbalance model of cervical syndrome through operation, so as to induce the degeneration of intervertebral disc. Rats began to receive the gastric perfusion from the 4^th month after operation, and rats in the Lingzhi group were given 400 mg/kg Ling:hi (Ling:hi spore powder was provided by Shanghai Lvgu Corporation), which was made into the suspension by dissolving in 10 mL water for gastric perfusion. Rats in the other two groups were administrated with same-volume water once a day for two months. The samples of C3-4,C4-5, C5-6 intervertebral disc were obtained from rats for histomorphologic observation. Meanwhile, ELASA was adopted to inspect the contents of IL-1β and TNF-α, and the content of NO was determined by optical density. Differences among groups were compared. RESULTS: A total of 24 experimental animals were involved in the analysis of results, and no one withdrew from the study. ①Observation under the microscope on the changes of cervical intervertebral disc in histomorphology at 6 months after the experiment: leakage could be seen in the annulus fibrosus disci intervcrtebralis of rats in the normal group, which were slightly disorder, and there were shrinkage in the vertebral pulp, and irregular hyperplasy could be seen in the cartilage. No normal tissues of intcrvertebral disc could be found in rats of the model group, they were taken place
