摘  要:目的:观察荞麦蛋白对阈上和阈下剂量戊巴比妥钠致小鼠睡眠实验和自发活动的影响。方法:实验于2005-07/08在赣南医学院药理学教研室完成。①选用健康成年昆明种雄性小鼠90只。②观察荞麦蛋白对小鼠自发活动的影响:选取小鼠30只,按随机抽签法分为3组,每组10只:低、高剂量荞麦蛋白组眼分别按3,6mg/g剂量腹腔注射荞麦蛋白(由天津科技大学生物工程学院植化教研室提供,临用前用生理盐水配成10g/L的荞麦蛋白溶液)演。对照组(腹腔注射等量生理盐水)。15min后放入吊笼中,稳定3min后采用LMS-2A型二道生理记录仪记录用药后15s内小鼠活动波形(次数)。③观察荞麦蛋白对腹腔注射阈上剂量的戊巴比妥钠小鼠睡眠功能的影响:选取小鼠30只,分组及各组干预措施同“自发活动实验”。末次给药15min后腹腔注射阈上剂量的戊巴比妥钠穴2.5g/L雪0.02mL/g。以小鼠翻正反射消失为入睡指标,从翻正反射消失至恢复的这段时间为睡眠持续时间。④观察荞麦蛋白对腹腔注射阈下剂量的戊巴比妥钠小鼠睡眠功能的影响:选取小鼠30只,分组及各组干预措施同“自发活动实验”。末次给药15min后腹腔注射阈下剂量的戊巴比妥钠穴2.5g/L雪0.01mL/g。比较3组入睡动物数、入睡时间和睡眠持续时间。⑤组间计量资料差异比较采用t检验。结果:小鼠90只均进入结果分析。①荞麦蛋白对小鼠自发活动的影响:给药后荞麦蛋白低和高剂量组二道生理记录仪记录15s内大波、中波出现个数明显少于对照组(P<0.05,0.01)。②荞麦蛋白对阈上剂量戊巴比妥钠诱发小鼠睡眠功能的影响:荞麦蛋白低和高剂量组小鼠入睡时间明显短于对照组(P<0.05,0.01),睡眠持续时间明显长于对照组(P<0.05,0.01);且荞麦蛋白高剂量组作用更为显著(P<0.01)。③荞麦蛋白对阈下剂量戊巴比妥钠诱发小鼠睡眠功能的影响:荞麦蛋白低和高剂�AIM: To observe the effect of buckwheat protein on the sleep and spontaneous motion of mice induced by pentobarbital sodium of suprethreshold dosage or subthreshold dosage. METHODS: The experiment was completed in the Department of Pharmacology, Gannan Medical University from February to March 2005.① Ninety healthy adult male mice of Kunming species were adopted. ②Effect of buckwheat protein on the spontaneous motion of mice: Thirty mice were selected to randomize into three groups with 10 rats in each: buckwheat protein groups of low-dose and high-dose (intraperitoneal injections of 3 mg/g and 6 mg/g buckwheat protein, provided by Department of Phytochemistry, College of Bioengineering, Tianjin .University of Science and Technology, and prepared into buckwheat protein solution with 10 g/L saline before use) and control group (Mice were injected intraperitoneally with distilled water of equal dosage), Every mouse was put into the suspended cage 15 minutes later, and stabilized for 3 minutes, then the wave pattern of mice's action was recorded with LMS-2A physiological grapher within 15 seconds after administration. ③Effect of buckwheat protein on the sleep of mice induced by pentobarbital sedium of suprathreshold dosage: Thirty mice were grouped and 'treated as above, After 15 minutes of final administration, pentobarbital sodium of suprathreshold dosage was injected intraperitoneally 42.5 g/L, 0.02 mL/g) into every mouse. The disappearance of fighting reflex of mice was taken as index of falling asleep, and the time from disappearance to rehabilitation of fighting reflex was recorded as sleep time. ④Effect of buckwheat protein on the sleep of mice induced by pentobarbital sodium of subthreshold, dosage: Thirty mice were grouped and treated as above.After 15 minutes of final administration, pentobarbital sodium of subthreshold dosage was injected intraperitoneally (2.5 g/L, 0.01 mL/g) into every mouse. Mice of three groups were compared in the amount of mice falling asleep,
