摘 要:目的研究小型猪的骨髓基质细胞(bone mesenchymal stem cell,BMSC)及其诱导后的成骨细胞在种植体钛合金(Ti-6Al-4V)表面的粘附情况。方法分离培养小型猪的骨髓基质细胞,并将其诱导为成骨细胞。分别将不同浓度的骨髓基质细胞及诱导后的成骨细胞悬液接种于种植材料表面,利用扫描电镜观察其粘附情况,计算不同接种浓度下两种细胞与种植材料的粘附率及粘附的细胞数。结果两种细胞在种植体表面粘附良好,接种浓度为3×105/100μl组两种细胞在种植材料表面的粘附细胞数量分别为17.6×106和18.6×106;粘附率分别84.2%和85.5%。两种细胞在钛合金表面的粘附率和粘附的细胞数与其余三组(1×105/100μl;2×105/100μl;4×105/100μl)相比差异均有显著性。结论小型猪BMSC在体外可经条件培养液诱导为成骨细胞,接种浓度为3×105/100μl组骨髓基质细胞及成骨细胞在种植材料表面有良好的粘附能力。Objective To study the bone mesenchymal stem cells and osteoblasts induced by the bone mesenchymal stem cells and their adhesion to the titanium implant (Ti-6Al-4V). Methods BMSC was isolated from mini pig bone marrow and induced to osteoblasts. Two kinds of cells with different density were seeded on the titanium implant. The number of cells adhered to the implant was counted and the adhesion rates were calculated. Results When cell density was up to 3 × 10^5/100μl the adhesion rates to the implant was 84.2 % and 85.5%. The number of cells adhered to the implant was 17.6× 10^6 and 18.6 × 10^6. The adhesion rate and the number of cells in the 3 × 10^5/100μl group were significantly higher than other groups. Conclusion The BMSC of the mini pig can be induced to osteoblasts in defined medium in vitro. When the seeding density is 3 × 10^5/100μl ,the bone mesenchymal stem cells and osteoblasts have good biocompatibility with the titanium implant.
