摘  要:目的:观察全反式维甲酸(ATRA)对人胰腺癌细胞株PC-3生长的抑制作用以及细胞株PC-3 Survivin基因表达的变化。方法:应用MTT比色法检测细胞增殖抑制率,RT-PCR法测定Survivin基因表达,流式细胞仪检测细胞凋亡。结果:经过ATRA作用后,PC-3细胞增殖受到抑制,呈时间和剂量依赖性。凋亡比例明显增加,Survivin mRNA的表达随着作用时间的延长而呈逐渐下降趋势。结论:ATRA对人胰腺癌细胞株PC-3的生长有抑制作用,同时可诱导其凋亡,提示其可能与Survivin基因表达下调有关。Objective: To observe the inhibitiory effects of all-trans retinoie acid (ATRA) on the pancreatic cancer cells growth in vitro and Survivin gene expression induced by ATRA, Methods: The inhibition rates of proliferation were detected by MTT assay. RT-PCR and flow cytometry were used to detect the expression of Survivin gene and the apoptotic rates. Results: After the treatment with ATRA, the proliferation of PC-3 was inhibited in dose-dependent and time-dependent manner,and the apoptotic rates of PC-3 cells were increased significantly. The expression of Survivin mRNA was decreased obviously in PC-3 cells receiving ATRA treatment compared with those receiving no treatment. Conclusion: ATRA can inhibit pancreatic cancer cells growth,induce cells apoptosis. Survivin gene probably contributes to the apoptosis of pancreatic cancer cells induced by ATRA.
