摘 要:目的:探讨大蒜新素(allitridin)在细胞水平对鼠肝炎病毒3型(mice hepatitis virus,MHV-3)的抑制作用。方法:细胞培养、蚀斑试验测定病毒滴度;MTT法进行药物毒性试验;蚀斑抑制试验检测大蒜新素的抗MHV-3活性,改变给药时间探索大蒜新素抗MHV-3作用环节。结果:大蒜新素半数中毒浓度(TC50)为75mg/L,抗MHV-3病毒的半数有效浓度(IC50)为2.7mg/L,治疗指数(TI)为27.78,大蒜新素对MHV-3病毒的抑制作用存在明显的量效反应关系(P<0.01),并且对MHV-3病毒有预防和中和作用(P<0.01)。结论:大蒜新素在L2细胞中对MHV-3病毒有明显的抑制作用,其作用机制是多途径的。Objective: To investigate the effect of allitridin injection on hepatitis virus strain 3(MHV-3) in mouse and the mechanism. Methods ;A MHV-3 infected cell model was established by a MHV-3-infected L2 cell cultures, The maximum tolerance concentration (MTC) of L2 cell bearing to allitridin treatment was measured with MTr colorimetric assay.The median effective inhibitory concentration (IC50) of allitridin was determined by plaque-reduction assay.The antiviral effect of allitridin was studied by using allitridin with different administration procedures and routes. Results :The median toxic concentration (TC50) of allitridin was 75 mg/L, and the IC50 of allitridin was 2.7 mg/L. The treatmen index (TI) was 27.78. The MTC of allitridin was 7.6 mg/L. No matter what routes of allitridin was used in,the MHV-3 was effectively suppressed by allitridin treatment. Conclusion :Allitridin is an effective anti-MHV- 3 medicine and has various antviral mechanism.
