摘　　要：目的探讨 SAMP10小鼠 NF-E2、YB-1、LRG47基因表达与脑衰老的相关性。方法采用快速老化小鼠 SAMP10、SAMR1为模型,运用 RT-PCR 和地高辛标记的非放射性 Northernblot 技术,观察8月龄 SAMR1对照组、SAMP10组前脑、皮质和海马 NF—E2、YB-1、LRG47基因表达的变化。结果 SAMP10组在前脑、皮质、海马中 NF-E2的表达(0.58±0.16、0.60±0.11、0.61±0.10)、YB-1的表达(0.48±0.13、0.47±0.11、0.46±0.10)、LRG47的表达(0.55±0.08、0.54±0.12、0.56±0.14)与 SAMR1对照组在前脑、皮质、海马中 NF-E2(0.36±0.09、0.4l±0.10、0.38±0.10)、YB-1(0.33±0.09、0.30±0.08、0.31±0.11)、LRG47(0.40±0.10、0.38±0.09、0.37±0.07)的表达比较,均明显下调(P<0.05)。结论 SAMP10小鼠脑衰老与转录调节因子 NF-E2、YB-1、LRG47基因表达异常有关。Objective To discuss the correlations of NF-E2, YB-1 and LRG47 gene expressions with brain senescence in senescence-accelerated mouse prone 10 （SAMP10） mice. Methods To measure the expressions of NF-E2, YB-1 and LRG47 genes in procerebrum, neocortex and hippocampus from a strain of SAMP10 and a strain of senescence-accelerated resistant mice （SAMR1） at age of 8 months by RT-PCR and DIG probed Northern blot techniques. Results The expressions of NF-E2 in procerebrum, neocortex and hippocampus （ 0. 58 ± 0.16,0.60 ± 0.11,0. 61 ± 0.10 ）, YB- 1 （0.48±0.13,0.47±0.12,0.46±0.10）and LRG47（0.55±0.08,0.54±0.12,0.56±0.14） were down regulated in SAMP10 group, when compared with SAMR1 control groups in the expressions of NF-E2 （0. 36 ± 0.09,0. 41±0.10,0.38±0.10）, YB-1 （0.33 ± 0.09,0.30 ± 0.08,0.31 ± 0.11） and LRG47（0.40±0. 10,0.38±0.08,0.37±0.07） （all P〈0.05）. Conclusions Brain senescence of SAMP10 mice is significantly associated with the abnormal expressions of gene NF-E2, YB-1 and LRG47.
