摘  要:目的:制备分泌特异性抗人甲状腺球蛋白(Tg)单克隆抗体的杂交瘤,建立检测Tg的ELISA方法。方法:以人甲状腺球蛋白为抗原免疫BALB/c小鼠,通过细胞融合制备分泌抗人TgmAb,并对其进行特异性鉴定,建立双抗体ELISA夹心法检测Tg。结果:共获得7株可稳定分泌抗人TgmAb的杂交瘤细胞株FMU-TG1~FMU-TG7,用单抗FMU-TG和Tg多克隆抗体建立了检测人Tg夹心ELISA试剂盒,敏感性可达30ng/ml,结论:成功制备了抗人TgmAb并建立了检测人Tg夹心ELISA试剂盒,对Tg的基础研究及临床应用具有重要意义。Objective: To prepare monoclonal antibody (mAb) against human thyroglobulin and establish the method of ELISA to detect human thyroglobulin. Methods: BALB/c mice were immunized by human thyroglohulin, the splenecytes of immunized mice were fused with Sp2/0 myeloma ceils by a routine method. The hybridoma ceils secreting specific antibody were detected . The stability of the obtained hybridoma ceils and the specificity of antithyroglobulin mAb the hybridoma ceils secreted were identified. Based all above, the double antibody sandwich ELISA method was established to detect thyroglobulin. Results:Seven hybridoma ceil lines which could secrete specific mAb stably were obtained( FMU -TG1 -FMU -TG7 ). A sandwich ELISA kit for detecting human thyroglobulin was established by using mAb FMU -TG and polyclonal antibody of thyroglobulin, and its sensitivity reached to 30ng/ml. Conclusion: The antithyroglobulin mAb was prepared and a sandwich ELISA kit for the detection of human thyroglobulin was estabhshed successfully.
