摘  要:目的利用携带SSTR2基因的重组腺病毒载体Adeno-X-SSTR2转染裸鼠胰腺癌皮下模型,使用SSTR2特异激动剂NC-8-12,观察其在体内实验中是否能取得抑制胰腺癌细胞增殖的效果。方法构建重组携带SSTR2基因的肿瘤增殖缺陷型腺病毒载体Adeno-X-SSTIL2,建立裸鼠胰腺癌皮下模型,分为转染和未转染SSTR2基因的两组,皮下分别给予不同剂量NC-8-12,观察肿瘤体积和重量的变化。结果在裸鼠皮下模型实验中。NC-8-12对未转染SSTR2基因的胰腺癌细胞无抑制作用(P>0.05);在转染了SSTR2基因的胰腺癌细胞中。NC-8-12在每日200μg/kg体重剂量下可以明显抑制肿瘤细胞增殖(P<0.05)。结论(1)携带SSTR2基因的肿瘤增殖缺陷型腺病毒载体Adeno-X-SSTR2可以高效转染胰腺癌细胞株。(2)一定剂量的NC-8-12(每日200μg/kg体重)对裸鼠体内转染了SSTR2基因的胰腺癌细胞的生长有明显抑制作用。Objective To observe whether it can inhibited the growth of pancreatic carcinoma cells in in vivo experiments with NC-8-12 (specific agonist of somatostatin receptor Ⅱ) by transfecting pancreatic carcinoma hypodermic model of naked rats with a recombinant adenovirus vector, named as Adenc-X-SSTR2, which carrying SSTR2 gene. Methods SSTR2 gene was cloned and recombined into Adeno-X-SSTR2 gene, constitute hypodermic model of naked rats with infecting and uninfecting SSTR2 gene separately, which were given NC-8-12 of various dose by hypodermic injection, to observe the change of tumor volume and weight by taking normol saline group as control group. Results NC-8-12 has no inhibition to pancreatic carcinoma ceils without transfecting SSTR2 gene in hypodermic model of naked rats (P〉0.05). But it can inhibit tumor ceils proliferation remarkably at the dose of 200μg/kg·d^-1 in the ceils transfected SSTR2 gene (P〈0.05).Conclusion (1) Adenc-X-SSTR2 with SSTR2 gene can infect pancreatic carcinoma ceils efficently. (2) a certain dose of NC-8-12 (200μg/kg·d^-1) can inhibited the growth of carcinoma ceils remarkably, which are transfected with SSTR2 gene in vivo models of naked rats.
