EGFR及C-erbB-2蛋白表达与腺样囊性癌细胞系转移力之间的关系

摘　　要：目的:研究表皮生长因子受体(EGFR)和C-erbB-2在涎腺腺样囊性癌2个细胞系中的表达,探讨其与腺样囊性癌肺转移的关系。方法:培养人涎腺腺样囊性癌SACC-83和肺高转移性腺样囊性癌SACC-LM细胞系,分别提取它们的胞质和胞膜样品并采用Western Blot技术检测EGFR和C-erbB-2在其中的表达差异,利用电泳凝胶成像分析软件对结果进行量化分析。结果:EGFR在SACC-83细胞的细胞膜中呈现高表达(P<0.01),而在SACC-LM细胞的胞质中呈现高表达(P<0.01)。SACC-83与SACC-LM细胞比较,前者胞膜中EGFR的表达明显高于后者,而胞质中的表达却明显低于后者(均为P<0.01)。C-erbB-2无论在SACC-LM细胞的胞质和胞膜中的表达都明显高于SACC-83细胞系,经灰度值分析,差异非常显著(P<0.01)。结论:EGFR基因在胞质中的高水平积累以及C-erbB-2基因的高水平表达可能在腺样囊性癌肺转移中发挥重要作用。Objective ：To investigate the expressions of epidermal growth factor receptor（ EGFR ） and C-erbB-2 in two salivary adenoid cystic carcinoma cell lines with different metastatic potential. Methods： Salivary adenoid cystic carcinoma SACC-83 cell line and its lung metastatic cell line SACC-LM were cultured in vitro, their cytoplasm and membrane samples were extracted, and the expressions of EGFR and C-erbB-2 were detected by Western blot, the results were quantitatively analyzed by FluorChem V2.0 software. Results ： EGFR expression level in cell membrane was higher than that in cytoplasm（ P〈0.01 ） in SACC-83 cell line and lower（ P〈0.01 ） in SACCLM cell line. It is higher in SACC-LM cytoplasm than in SACC-83 cytoplasm（ P 〈0.01 ） and lower in SACC-LM cell membrane than in SACC-83 cell membrane（ P〈0.01 ）. C-erbB-2 expression levels in SACC-LM cytoplasm and cell membrane were both higher than those in SACC-83 （P〈0.01 ）. The expression of C-erbB-2 in cytoplasm was not significantly different from that in cell membrane（ P〉0.05 ） in both cell lines. Conclusion：Higher EGFR expression in cytoplasm and higher C-erbB-2 expression may be related to the metastatic potential of SACC-LM cells.
