热休克蛋白70和90α在人胃癌组织中表达及其与胃癌生物学行为的关系

摘　　要：目的:研究热休克蛋白70和90α在人胃癌组织和正常黏膜组织中的表达,探讨其表达与胃癌生物学行为的关系。方法:41例胃癌组织和相应的正常黏膜组织分别行免疫组织化学染色和RT-PCR检测,分析检测结果。结果:热休克蛋白70和90α在胃癌组织中表达分别为75·6%(31/41)和80·5%(33/41),明显高于正常黏膜组织中的表达,在分化较差的肿瘤类型中表达分别为85·2%和88·9%,高于分化较好的肿瘤类型,在淋巴结转移阳性的病例中分别为82·8%和86·2%,高于淋巴结转移阴性的病例,在TNM分期Ⅲ、Ⅳ期的病例中表达分别为80·6%和83·9%,高于其他分期。结论:热休克蛋白70和90α在胃癌组织中明显表达升高,且在晚期的病例中表达高于相对较早期的病例,与胃癌的生物学行为有关。OBJECTIVE：To assess the expressions of heat shock protein 70 and heat shock protein 90α in gastric cancer and normal mucosa, and to determine their relation to biological behaviors of gastric carcinoma. METHODS： Immunohistochemical staining and RT-PCR were performed in 41 cases of gastric cancer and normal mucosa for the expressions of HSP70 and 90α, and the results were analysised. RESULTS： The positive expression rates of HSP70 and HSP90α were 75.6 % （31/41） and 80.5 % （ 33/41 ） in gastric cancer, respectively, they were higher than ones in mucosa, The positive expression rates of HSP70 and 90α in poor differentiation cancer were 85.2 % and 88. 9 %, respectively, higher than ones in good differentiation cancer. The posi- tive expression rates of them in gastric cancer with lymph node metaste- sis were 82.8% and 86.2%, respectively, higher than ones in gastric cancer without lymph node metastesis. The positive expression rates of them in l]I, IV stage were 80.6% and 83.9%, respectively, higher than ones in other stages. CONCLUSIONS： The expressions of HSP70 arid HSP90α increase in gastric cancer, and they were higher in advanced stage tumor than ones in early stage tumor, and they were closely related to the biological behaviors of gastric carcinoma.
