TJU103联合CTLA4-Ig对T细胞增殖和白血病细胞毒活性的影响

摘　　要：目的:观察异烟腙类衍生物TJU103联合CTLA4Ig对供者T细胞增殖和白血病细胞毒活性的影响。方法:体外建立HLA半相合供、受者间单向混合淋巴细胞反应(mixedlymphocytereaction,MLR)体系,应用TJU103联合CTLA4Ig阻断供者T细胞激活的抗原和共刺激双信号通路,利用MTT法检测供者T细胞经受者正常抗原刺激下的增殖反应以及对受者原代白血病细胞的杀伤活性。结果:单独应用TJU103和CTLA4Ig均能够降低供者T细胞的增殖活性,两者联合应用抑制增殖效应明显增强,增殖抑制率达92%,优于单独应用组(P<0.05),但不影响供者T细胞对原代白血病细胞的杀伤活性,P>0.05。结论:TJU103联合CTLA4Ig能够阻断供者T细胞激活的抗原和共刺激双信号通路,在明显降低供者T细胞增殖反应的同时,不影响其细胞毒活性的发挥。OBJECTIVE： To investigate the proliferation and cytotoxicity of donor T cells with a combination of TJU103 and CTLA4-Ig METHODS： MTT assays were used respectively for detection of T lymphocytes proliferation and its anti-leukemia effects after given a combination of TJU103 and CTLA4-Ig in MLR. RESULTS： There were significant differences in the prohibition of T-cell proliferation between groups of TJU103, CTLA4-Ig alone or combined group of TJU103 and CTLA4-Ig （P〈0.05）, but there were no differences in the ability of anti-leukemia between each groups （P〈0.05）. CONCLUSION： The combination of TJU103 and CTLA4-Ig could significantly prohibit the T-cell proliferation but the ability of anti-leukemia is not influenced.
