交联型异体脱细胞硬脑膜行眼部修复的形态学及抗原性研究

摘　　要：目的将交联型异体脱细胞硬脑膜植入眼部,研究其组织相容性及抗原性变化。方法将20只新西兰大白兔随机平均分为两组,去除睑板后,分别植入兔交联型脱细胞硬脑膜和异体硬脑膜,于植入后1、4、8周观察全身免疫学指标的变化;并行光镜和免疫组织化学检查。结果脱细胞硬脑膜植入眼部后炎症反应轻,外周血IgA、IgM、C3和CD4+T细胞、CD8+T细胞的改变无明显差异,脱细胞组外周血IgG(术后1周)和植入局部的CD4+T细胞、CD8+T细胞浸润明显少于异体硬脑膜组。结论脱细胞硬脑膜植入眼睑后引起的免疫反应较异体硬脑膜小。Objective The less inflammatory reaction and good histocompatibility make the implantation of tissue regeneration materials to be a ideal repairing and reconstructing way of eyelid. Acellular duramater is the one of these materials. This paper was to find a method of preparation of acellular duramater and investigate the histocompatibility of acellular dura implantation in rabbit eyelid. Methods Acellular dura was prepared with 2% Triton X-100, DNase and RNase. Twenty New-Zealand white rabbits（20 right eyes）were divided into two groups randomly. Dura was implanted the big tarsus with 5 mm ×8 mm in upper eyelid in dura group and acellular dura was done at the same operation in acellular dura group. The appearances, immunologic changes and inflammatory reaction of eyelids after operation were observed in 1, 4, 8, 12 weeks in postoperation with HE stain and immunofluorescence assay. Results Acelullar dura could be successful prepared with 2% Triton X-100. There were no statistical differences between two groups in IgA,IgM,C3 ,CD^4＋T cells and CD^8 ＋T cells of blood（P 〉0. 05）. But the IgG level of blood was different 1 week in postoperation（P 〈 0. 05 ）. In the 4th week, the average numbers of CD^4＋ T cells and CD^8 ＋ T cells in eyelid of rabbit in dura group were 31 ± 4 and 23 ± 5, while 　those of acelllar dura group were 9 ± 4 and 8 ± 2 without significant difference between two groups. Conclusion Acellular dura is easy to be prepared and could be used to the repairing of eyelid.
