摘  要:目的研究肝硬化大鼠胃窦平滑肌细胞色素氧化酶Ⅱ基因(COⅡgene)及线粒体膜电位的变化,探讨肝硬化胃动力障碍机制。方法20只Wister大鼠随机分为肝硬化组和对照组,采用葡聚糖蓝-2000为胃肠内标记物观察胃动力的变化,RT-PCR法检测胃窦平滑肌COⅡmRNA的变化,激光共聚焦显微镜检测线粒体膜电位的改变。结果与对照组比较,肝硬化组胃动力明显减弱(P<0.01),胃窦平滑肌COⅡmRNA的表达及线粒体膜电位均显著低于对照组(P<0.01)。结论肝硬化大鼠胃运动功能减退与胃窦平滑肌COⅡmRNA的表达下降及线粒体膜电位的降低有关。Objective To study the changes of cytochrome C oxidase gene Ⅱ mRNA(CO Ⅱ mRNA) and mitochondrial membrane potential(MMP) in smooth muscle cells of antrum in liver cirrhosis rats. Methods 20 Wister rats were randomly divided into control group and liver cirrhosis model group, the CO Ⅱ mRNA in antral smooth muscle cells was assayed with reverse-transcriptional polymerase chain reaction(RT-PCR) and mitochondrial membrane potential was detected by laser scanning confocal microscope. The gastric motility of rats were observed through Dextran blue-2000 indicator. Results Comparing with control group, both the expression of CO Ⅱ mRNA and mitochondrial membrane potential were significantly decreased in model group[CO Ⅱ mRNA(0. 51±0. 06) vs (1.13±0.09) D; MMP(269.32 ±67.13) vs (864.53± 157.42),P〈0. 017, and the gastric motility of rats was markedly delayed(P〈 0.01). Conclusion The delaying gastric motility of rats with cirrhosis may be associated with the low expression of CO Ⅱ mRNA and the decrease of mitochondrial membrane potential.
