摘 要:目的建立一种可以测定动物组织中自由基含量的化学方法。方法取老龄小鼠心、肝、脾、肺、肾、脑、肌肉的组织样品,以及Beagle犬口服超氧化物歧化酶(SoD)溶液以后不同时间的血浆样品,用脂质过氧化法测定其中自由基的含量。结果标准曲线回归方程为:A=0.0285 C-0.0443(r=0.9970),线性范围为8.27—41.35nmol·ml^-1,日内和日间精密度RSD均低于8%,高、中、低浓度样品的加样回收率分别为100.68%、99.66%、97.95%。测得小鼠体内各组织的自由基含量和分布与文献报道的相符合。而Beagle犬口服SOD溶液后血浆中的自由基含量逐渐降低,并于2.5h出现最低值。结论脂质过氧化法测定动物体内自由基的含量,线性和重复性均好,加样回收率高,仪器简单,试剂价廉,操作方便。OBJECTIVE To establish a new method, called lipid peroxidation method, to determine the free radicals in vivo. METHODS The standard curve was established with precision and recovery rates being determined. The hearts, livers, spleens, lungs, brains, muscles of aged mice, and plasma of Beagle dog after given SOD solution, were determined by lipid peroxidation method. RESULTS The regression equation was A = 0. ff285 C - 0. 0443 ( r = 0.9970 ), with linear range of 8.27 being 41.35 nmol· ml^- 1. RSD was lower than 8 %, and the recovery rates of added samples being 1130.68 %, 99.66 % and 97.95 %, respectively. The contents of free radicals determined in various tissues of aged mice accord with literature, and free radicals in plasma of Beagle dog decrease with time and reach the lowest value at 2.5 h after given SOD solution. CONCLUSION The method is simply equiped, cheap and conveniently operated, with good linearty and precision.
