摘 要:目的探讨抑癌基因NORE1A在肺癌组织中的表达缺失情况及其临床意义。方法采用RT-PCR技术检测46例肺癌组织及相应癌旁正常组织中NORE1A基因的表达缺失情况。结果(1)NORE1A基因在正常肺组织均有表达,在肺癌组织中表达缺失率为45%;(2)NORE1A基因在不同病理细胞类型标本中的表达缺失率不存在明显差异,P>0.05;(3)吸烟组和非吸烟组间NORE1A基因的表达缺失比较差异无显著性,吸烟组缺失率为48%,非吸烟组为42%(P>0.05);(4)淋巴结转移组的NORE1A基因的表达缺失率高于无淋巴结转移组标本,分别为60%和18%(P<0.01)。结论不能认为吸烟是导致NORE1A基因失活的一个具有重要影响的因素。NORE1A基因的表达缺失与肺癌发生有关,不仅发生于早期肺癌,而且可能在其晚期亦存在继发性大量失活。Objective To explore the expression loss of NORE1A gene transcripts and clinical significance in lung carcinoma. Method The mRNA expression of NORE1A was detected by RT-PCR in 46 human lung cancer tissues and their matched normal tissues. Results ( 1 ) The expression of NORE 1A gene transcripts was detectable in all normal lu.ng tissues, but undetectable in some lung cancer tissues. The rate of loss in lung cancer tissues is 45%. (2)There was no significant association about the rate of expression loss of NORE1A transcripts in all collected different histological types, (P 〉 0.05 ). (3) No significant association about the rate of expression loss of NORE1A transcripts was detected between the smoking group and no-smoking group, the rate of expression loss in the smoking group is 48% ,the rate of expression loss in the no-smoking group is 42 %, χ^2 = 0.122, P = 0.727 ( P 〉 0.05 ). (4) The rate of loss of NORE1A transcripts was significantly higher in the tissues with positive lymph node metastasis than in those without lymph node metastasis. The rate of loss in the tissues with positive lymph node metastasis is 60% ,the rate of loss in those without lymph node metastasis is 18%, (χ^2 = 7. 156 ,P = 0.007 ). Conclusion Smoking should not be regarded as a significant influence factor resulting in the expression loss of NORE1A transcripts, and not only occurred the expression loss of NORE1A gene in early stage lung cancer,but is also continuous a great quantity loss of NORE1A gene in terminal lung cancer.
