摘  要:目的探讨实验性变态反应性脑脊髓炎(EAE)T淋巴细胞增殖和肿瘤坏死因子(TNF)α变化的意义。方法从豚鼠(GP)脊髓中分离髓鞘碱性蛋白(MBP),主动免疫Lewis大鼠诱导EAE成功后,观察EAE组临床症状,并进行临床评分;应用淋巴细胞增殖试验测定EAE大鼠淋巴结T淋巴细胞增殖程度;应用ELISA技术测定血清TNFα水平。结果EAE临床发病过程中,T淋巴细胞增殖活跃,血清TNFα水平异常升高,并与临床症状呈正相关。结论EAE时T淋巴细胞增殖旺盛和TNFα生成增多,进而导致血脑屏障(BBB)的破坏,炎细胞浸润,从而诱发EAE。Objective To explore the relationship between T cell proliferation, serum tumor necrosis factor (TNF)-α abnormalities and clinical course of experimental autoimmune encephalomyelitis (EAE) in Lewis rats. Methods The rats of EAE were examined for the development of neurological signs of disease, T cell proliferation responses and TNF-α concentrations. Results There was an active T cell proliferation, and a marked TNF-α increase which paralleled with clinical onset in EAE. Conclusion Our studies suggest that T cell proliferation and increasing of TNF-α result in blood-brain barrier (BBB) breakdown and significant inflammatory cell recruitment which leads to EAE.
