摘 要:为了探讨运动对长骨发育的作用机理,采用反转录聚合酶链反应(RT-PCR)技术,对100只生长期大鼠经3周、6周和9周的高、低负荷跑台训练后胫骨生长板中局部调节因子甲状旁腺素相关蛋白(PTHrP)的表达进行了观察。结果表明:3周的跑台运动没有显著改变生长板中PTHrP mRNA的表达水平,而6周和9周运动后PTHrP mRNA的表达水平显著高于对照组,低负荷运动组亦显著高于高负荷运动组(P<0.01或0.05);各组间对照组PTHrP mRNA的表达水平均未出现显著的增龄变化。结果提示:运动可以通过改变PTHrP的表达水平来调节生长板软骨细胞的活动,进而影响生长板的高度,调节长骨的纵向生长,但其具体的调节途径尚有待进一步研究探讨。In order to explore the mechanism of the effect of exercise on the longitudinal development of the long bone, 100 male SD growing rats were randomly divided into three groups (baseline control, sedentary control and exercise group) and ten subgroups, and their expression of the Parathyriod Hormone-Related Protein (PTHrP) in growth plate were investigated by reverse transcription polymerase chain reaction technique ( RT-PCR). The results showed that the level of PTHrP mRNA in 3 weeks exercise group weren' t changed, and there were significant difference between exercise and control groups as well as low loading and high loading exercise groups in 6 and 9 weeks ( P 〈 0.01 or P 〈 0.05). However, there were no changes with age in each sedentary control group. The present study indicated that exercise could regulate the activity of chondrocytes in growth plate by changing the expression of PTHrP mRNA, further affect the height of growth plate. But the pathway that different intensive exercises regulate the longitudinal growth of the long bone by changing the expression of PTHrP should be further investigated.
