摘 要:目的:通过检测荷肝癌小鼠射频治疗后脾细胞分泌Th1类细胞因子IL-2、IFN-γ及Th2类细胞因子IL-4、IL-10的动态变化,评价射频热疗对小鼠免疫状态的影响。方法:用酶联免疫吸附(ELISA)方法检测荷瘤组、手术组、射频组、正常对照组小鼠脾细胞培养上清IL-2、IFN-γ、IL-4、IL-10的表达量。结果:IL-2含量在射频治疗后2周显著高于正常组2周和手术后2周(P<0·05);IFN-γ含量在射频后1、2、3周均显著高于手术后1、2、3周及正常对照组1、2、3周(P<0·05);IL-4含量在射频后1、2、3周均显著低于同期手术后和正常对照组(P<0·05);IL-10含量在射频治疗后2周亦显著低于正常组2周和手术后2周(P<0·05);而同期手术组和正常对照组间无显著差异(P>0·05)。结论:射频治疗可以促进Th1类细胞因子IL-2和IFN-γ的表达,使荷瘤小鼠由Th2向Th1逆转,从而增强机体的抗肿瘤免疫功能。AIM: To evaluate the influence of radiofrequeney (BF) hyperthennia on immunity function in mice. METHODS: The expression pattern of T helper type 1 (Thl) and T helper type 2 (Th2) cytokines in splenocyte culture supernatants, mainly the expression levels of IFN-γ, IL- 2, IL-4 and IL- 10 in splenocyte culture supernatants of mice in tumorbearing group, surgical resection group, BF therapy group and normal control group were detected by enzyme- linked immunoadsordent assay (FLISA). RESULTS: IL- 2 concentration in two weeks after RF therapy group was higher than that in two weeks after smgical resection and normal control groups ( P 〈 0.05). IFN -γ concentration in one week, two weeks and three weeks after RF therapy groups was higher than that in one week, two weeks and three weeks after statical resection and normal control groups, respectively ( P 〈 0.05). IL - 4 concentration in one week, two weeks and three weeks after RF therapy groups was lower than that in one week, two weeks and three weeks after statical resection and normal control groups, respectively ( P 〈0.05). IL- 10 concentration in two weeks after RF therapy group was lower than that in two weeks after statical resection and normal control group ( P 〈 0.05). It had no significant difference in surgical resection groups and normal control groups ( P 〉 0.05). CONCLUSION: RF hyperthermia may activate the transformation from Th2 to Thl and facilitate the excretion of Thl type cytokines that play an important role in the anti- tumor immunity.
