摘  要:[目的]观察加味左金丸对大鼠胃癌前病变增殖细胞核抗原(PCNA,用增殖指数PI表示)和端粒酶活性(用hTERT表示)的影响。[方法]采用甲硝基亚硝基胍(MNNG)、0.03%雷尼替丁、56℃15%盐水、0.85%脱氧胆酸钠、40%乙醇、饥饱失常等多种因素制作大鼠胃癌前病变模型和免疫组化方法检测正常组、自然恢复组、加味左金丸高、中、低剂量组、维甲酸组大鼠胃黏膜组织中PCNA(换算成增殖指数PI)和hTERT蛋白表达情况。[结果](1)加味左金丸高、中、低组PI与维甲酸组比较差异有统计学意义(P<0.05),这四组均与自然恢复组比较差异有统计学意义(P<0.01或0.05)。(2)加味左金丸各组及维甲酸组hTERT蛋白阳性率均较自然恢复组降低,但仅中剂量组有统计学意义(P<0.05)。[结论]加味左金丸通过抑制端粒酶及细胞增殖活性,发挥逆转胃癌前病变的作用。[Purpose] To observe the effect of adjusted ZuoJin pills (AZP) on proliferating cell nuclear antigen (PCNA, represented by proliferating index, PI) and telomerase activity (represented by human telomerase reverse transcriptase, hTERT) in rats with gastric precancerous lesion (GPL). [Methods] The model was established by MNNG together with other factors including 15% 56℃ brine, sodium deoxycholate, 40% alcohol stimulus, 0.03% ranitidine, undue hunger, overeating. PCNA (converted to PI) and hTERT was detected by immunohistochemical method, in normal group, sponsaneous recovery group, AZP groups of high-dosage, medium-dosage, low-dosage, and retinoic (RA) group. [Results] (1) PI was significantly different between high, medium, and low-dosage AZP groups in each and RA group (P〈0.05). Also, PI was significantly different between above 4 groups in each sponsaneous recovery group (P〈0.01 or 0.05). (2) The positive expression rate of hTERT protein could be decreased in all the AZP groups and the RA group. There was significant difference only between the medium-dosage AZP group and the sponsaneous recovery group (P〈0.05). [Conclusion] AZP makes GPL reversed in rates by inhibiting activity of telomerase and cell proliferating.
