摘  要:目的观察γ-干扰素(interferon-γ,IFN-γ)对肿瘤坏死因子-α(tumornecrosisfactor-α,TNF-α)促正常人颊黏膜成纤维细胞(normalmucousfibroblasts,NM-FB)及口腔黏膜下纤维化(oralsub-mucousfibrosis,OSF)患者颊黏膜成纤维细胞(OSF-FB)胶原合成的影响,探讨IFN-γ用于治疗OSF的可能性。方法取6例OSF患者及5例正常人颊黏膜,行成纤维细胞原代培养并传代,采用羟脯氨酸试剂盒法观察IFN-γ对TNF-α作用下NM-FB及OSF-FB的胶原合成的影响,并在电镜下观察其超微结构的改变。结果IFN-γ对TNF-α促NM-FB及OSF-FB的胶原合成有明显抑制作用。结论IFN-γ是OSF形成过程中的一种负性调节因子,有可能用于临床治疗OSF。[Objective] To investigate the inhibition effects of IFN-γ on TNF-α promoted collagen biosynthesis of oral submucous fibrosis(OSF)-derived fibroblasts. [Methods] Fibroblast cultures were established from the buccal mucosa of five normal persons and six patients with OSF. The study focused on the effects of IFN-γ on the collagen biosynthesis of fibroblasts by TNF-α promoted in vitro. Also the changes of the ultrastructure were observed by eleetronmieroseopy. [Results] The study showed that IFN-γ could reduce the TNF-α promoted collagen biosynthesis of oral mucosal-derived fibroblasts. [Conclusions] The investigation confirm that IFN-γ is a cytokine with down-regulation effects on OSF progression, and suggest that IFN-γ can play an important role in prevention and treatment of OSF.
