摘 要:目的:观察家兔肢体缺血再灌注时凝血和纤溶系统活性的变化及葛根素对其的影响并探讨其机制。方法:健康家兔40只,随机分为四组:Ⅰ组为假手术对照组,Ⅱ组为缺血再灌注组,Ⅲ组为葛根素低剂量治疗组,Ⅳ组为葛根素高剂量治疗组。检测血浆超氧化物歧化酶(SOD)、丙二醛(MDA)、组织纤溶酶原激活物(tPA)、纤溶酶原激活物抑制物(PAI)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)的动态变化。结果:与缺血前和缺血4h比较,Ⅱ组在再灌注后tPA活性明显降低、APTT缩短,再灌注4h,PAI活性明显升高,TT缩短;Ⅲ组在再灌注后2h和4h时tPA活性明显降低,Ⅲ组和Ⅳ组各时间点之间PAI、APTT、PT及Ⅳ组各时间点之间tPA比较均无显著性差异。SOD活性与tPA活性正相关,MDA含量与tPA活性负相关。结论:肢体缺血再灌注使机体凝血功能增强,纤溶功能抑制,葛根素对凝血和纤溶功能异常有明显的改善作用,其机制与抗氧自由基,保护内皮功能有关。Objective : To investigate the Changes of blood eoagulative and fibrinolytic activities in rabbits after limb ischemia reperfusion and the effects of puerarin on them. Methods : 40 rabbits were randomly divided into 4 groups. Group Ⅰ were sham -operated animals; group Ⅱ, nontreated iachemia reperfusion animals; group Ⅲ, 40 mg/kg puerarin treated animals; and group Ⅳ, 60 mg/kg puerafin treated animals. Levels of blood SOD, MDA, tPA, PAI, PT and APTr were determined in all rabbits and the dynamic changes were observed. Results : In group Ⅱ, the activities of tPA after rcpeffusion were obviously lower than those before ischemia and after 4 hours of ischemia and the APTr were shortened. Similarly, the activities of PAl were greatly increased and the Tr were shortened after 4 hours of reperfusion. In group Ⅲ, the activities of tPA after2,4 hours of reperfnsion were obviously lower than those before isehemia and after 4 hours of ischemia. Levels of blood PAI, PT and APTr in different stages in group Ⅲ or Ⅳ showed no marked diversity, Nor it was with the activities of tPA in group Ⅳ. Conclusion: The function of blood coagulation was enhanced and the fibrinolytic activities were inhibited in rabbits after llmb ischemia reperfusion. Puerarin could greatly meliorate the abnormity of blood coagulafive and fibrinolytic functions after limb iachemia repcrfusion and the mechanism was concerned with its anti - oxidant activities and protecting the function of endotheliocyte.
