柔红霉素诱导Jurkat细胞凋亡的实验研究

摘　　要：目的探讨柔红霉素在体外诱导Jurkat细胞发生凋亡的规律.方法应用DNR体外作用于Jurkat细胞,研究DNR诱导Jurkat 细胞凋亡的规律,Annexin V/PI双标法流式细胞仪检测Jurkat细胞凋亡率,PI染色流式细胞仪检测细胞周期分布.结果 0.1～2.0μmol/L浓度范围DNR作用一定时间后,Jurkat细胞的凋亡率随药物浓度的增加与作用时间的延长而升高.DNR作用后Jurkat细胞G2/M期比例明显增加.结论一定浓度的DNR在体外可诱导Jurkat细胞凋亡,在一定的剂量范围内,呈现剂量-效应、时间-效应关系,其机制可能与DNR抑制细胞有丝分裂有关.Objective To study the rule of apoptosis of Jurkat induced by daunorubicin（DNR）. Methods Apoptosis of Jurkat cells stained with Annexin V/PI was detected by flow cytometry. Cell cycle distribution of Jurkat cells was analyzed by flow cytometry after cell stained with PI. Results The increase of apoptotic Jurkat cells percentage was in direct proportion to DNR doses within 0. 1-2.0μmol/L as well as exposure period. Jurkat cells exposed to DNR were demonstrated increased in the G2/M phase. Conclusion Apoptosis of Jurkat cells induced by DNR could be related to the inhibition mitosis of Jurkat cells.
