蛋白激酶C同工酶在乳腺癌中表达的研究

摘　　要：目的观察蛋白激酶C(prote in k inase C,PKC)同工酶PKCα-、PKCι-在乳腺癌组织中的表达状况,探讨癌细胞信息传递的分子生物学机制。方法应用半定量RT-PCR检测31例乳腺癌组织与12例癌旁正常乳腺组织中PKCα-、PKC-ιmRNA的表达;应用W estern b lot检测以上组织中PKCα-、PKC-ι蛋白表达。结果乳腺癌组织PKC-αmRNA与蛋白表达均高于癌旁正常乳腺组织(P<0.01);乳腺癌组织PKC-ιmRNA及蛋白表达与癌旁正常乳腺组织比较差异均无显著性(P>0.05)。结论乳腺癌组织细胞增殖可能与PKC-α同工酶过表达所介导的信号转导有关,与PKCι-同工酶无明显关系。Objective To investigate the expression pattern of protein kinase C （PKC） isoenzymes in breast cancer. Methods The expression of PKC -α and PKC-t was detected by semiquantitative RT-PCR and Western blotting in 31 cases of breast cancer and 12 cases of normal tissues. Results PKC-α mRNA and protein were markedly increased in cancer tissues compared with normal tissues（P〈0. 01）. There was no difference in the amount of PKC-t mRNA and protein between the cancer and normal tissues （P 〉 0. 05）. Conclusion PKC-α, not PKC-t plays an important role in regulating cellular proliferation of breast cancer.
