摘　　要：目的 研究金雀异黄素（GEN）对人结癌细胞系（SW480）细胞的作用并探讨其作用机理。方法 采用细胞计数法MTT法、免疫组化等方法，观察GEN对体外培养的SW480细胞的生长抑制以覆COX2表达的影响。结果 GEN对体外培养SW480细胞具有明显的生长抑制效应且呈剂量-效应的依赖关系，当50μmol／LGEN作用SW480细胞24h，抑制率达28％。免疫组化的结果显示：GEN处理后的SW480细胞COX2表达下降。结论 CEN对体外培养SW480细胞具有增殖抑制作用，CEN通过下调SW480细胞COX2蛋白的表达，促进细胞凋亡。Objective To explore the potential therapeutic effect of GEN on human colorectal cancer cell line SW480. Methods MTT was used to test the cell growth of SW480, and immunohistochemical staining was used to detect the expression of COX2. Results GEN significantly inhibited proliferation of SW480 cell and the inhibiting effects had dose - and- time - dependence. The inhibition fraction of SW480 cell exposed to 50μmol/L GFN for 24h was 28%, immunohistochemical staining revealed that the expression of COX2 SW480 cell reduced. Conclusion GEN can effectively inhibit the proliferation of human colorectal cancer cell lines SW480. GEN induce apoptosis in SW480 cells via downregulating the expression levels of COX2.
